Abstract

In this study, we investigated the effects of grape seed proanthocyanidin extract (GSPE) against the side effects of high-dose administration of methylprednisolone (MP) in male rats. A total of 32 adult Wistar male albino rats were divided into four groups: (1) control (CON), received standard food only; (2) MP, received standard food + intraperitoneal injection of 60 mg/kg MP on day 7; (3) GSPE, received standard food + 200 mg/kg/day GSPE; and (4) MP + GSPE, received standard food + 200 mg/kg/day of GSPE + intraperitoneal injection of 60 mg/kg MP on day 7. All animals in the GSPE and GSPE + MP groups were treated once a day by oral gavage for 14 consecutive days. The feed intake of rats in the MP and MP + GSPE groups decreased significantly by 24.14% and 13.52%, respectively ( < 0.05). Administration of MP resulted in significant increases in serum concentrations of blood urea nitrogen ( < 0.001), glucose ( < 0.01), alkaline phosphatase, and adrenocorticotropic hormone ( < 0.05). High-dose MP administration significantly reduced catalase ( < 0.001) and glutathione peroxidase ( < 0.05) concentrations in the liver and kidney tissues of rats, while glutathione concentrations were only reduced in liver tissue ( < 0.05). The expression levels of and in liver, kidney, and testicular tissue were significantly increased, while the expression levels of were reduced ( < 0.001). Furthermore, sperm concentration was significantly affected by GSPE in rats induced by high-dose MP, and sperm loss was significantly reduced in MP + GSPE ( < 0.05). These findings suggest that GSPE could be useful as a supplement to alleviate MP-induced toxicity in rats.

在本研究中,我们调查了葡萄籽原花青素提取物(GSPE)对雄性大鼠高剂量甲基强的松龙(MP)给药副作用的影响。总共32只成年Wistar雄性白化大鼠被分为四组:(1)对照组(CON),仅接受标准食物;(2)MP组,接受标准食物+在第7天腹腔注射60mg/kg MP;(3)GSPE组,接受标准食物+200mg/kg/天GSPE;(4)MP+GSPE组,接受标准食物+200mg/kg/天GSPE+在第7天腹腔注射60mg/kg MP。GSPE组和GSPE+MP组的所有动物连续14天每天经口灌胃给药一次。MP组和MP+GSPE组大鼠的采食量分别显著下降了24.14%和13.52%(P<0.05)。MP给药导致血清尿素氮(P<0.001)、葡萄糖(P<0.01)、碱性磷酸酶和促肾上腺皮质激素(P<0.05)浓度显著升高。高剂量MP给药显著降低了大鼠肝脏和肾脏组织中过氧化氢酶(P<0.001)和谷胱甘肽过氧化物酶(P<0.05)的浓度,而谷胱甘肽浓度仅在肝脏组织中降低(P<0.05)。肝脏、肾脏和睾丸组织中[具体基因名称1]和[具体基因名称2]的表达水平显著升高,而[具体基因名称3]的表达水平降低(P<0.001)。此外,高剂量MP诱导的大鼠精子浓度受到GSPE的显著影响,MP+GSPE组精子损失显著减少(P<0.05)。这些发现表明,GSPE可用作一种补充剂来减轻MP诱导的大鼠毒性。
