Abstract

Sulfoxy radicals (SORs) are oxygen- and sulfur-containing species such as SO, SO, and SO. They can be physiologically generated by S(IV) autoxidation with transition metal catalysis. Due to their harmful effects, the detection of both SORs and their scavengers are important. Here, a simple and cost-effective method for the determination of SORs and the scavenging activity of different antioxidant compounds was proposed. A SOR was selectively generated by combining CoSO·7HO with NaSO. To detect SOR species as a whole, 3,3',5,5'-tetramethylbenzidine (TMB) was used as the chromogenic reagent, where SOR generated in the medium caused the formation of a blue-colored diimine from TMB. Additionally, the SOR scavenging effects of a number of antioxidant compounds (AOx) belonging to different classes were investigated, among which catechin derivatives were the most effective scavengers. The obtained results were compared with those of a reference rhodamine B decolorization assay. The radical scavenging effects of the tested AOx were ranked by both assays and then compared using the Spearman statistical test to yield a very strong correlation between the two rankings. The method was applied to real samples such as catechin-rich tea, that is, white, black, and green tea, among which white tea was determined as the most effective SOR scavenger.

亚磺酰基自由基(SORs)是含硫和氧的物种,如SO、SO和SO。它们可通过过渡金属催化的S(IV)自氧化在生理条件下产生。由于其有害影响,检测SORs及其清除剂都很重要。在此,提出了一种简单且经济高效的方法来测定SORs以及不同抗氧化化合物的清除活性。通过将CoSO·7HO与NaSO混合选择性地生成SOR。为了整体检测SOR物种,使用3,3',5,5'-四甲基联苯胺(TMB)作为显色剂,其中在介质中产生的SOR会使TMB形成蓝色二亚胺。此外,研究了多种不同类别的抗氧化化合物(AOx)对SOR的清除作用,其中儿茶素衍生物是最有效的清除剂。将所得结果与参考罗丹明B脱色测定法的结果进行比较。通过两种测定法对测试的AOx的自由基清除作用进行排名,然后使用Spearman统计检验进行比较,以得出两种排名之间非常强的相关性。该方法应用于富含儿茶素的茶叶等实际样品,即白茶、红茶和绿茶,其中白茶被确定为最有效的SOR清除剂。
