Cerebral ischemia/reperfusion injury (CIRI) is a major contributor to poor prognosis of ischemic stroke. Flavonoids are a broad family of plant polyphenols which are abundant in traditional Chinese medicine (TCM) and have beneficial effects on several diseases including ischemic stroke. Accumulating studies have indicated that flavonoids derived from herbal TCM are effective in alleviating CIRI after ischemic stroke or , and exhibit favourable therapeutical potential. Herein, we systematically review the classification, metabolic absorption, neuroprotective efficacy, and mechanisms of TCM flavonoids against CIRI. The literature suggest that flavonoids exert potential medicinal functions including suppressing excitotoxicity, Ca overloading, oxidative stress, inflammation, thrombin's cellular toxicity, different types of programmed cell deaths, and protecting the blood-brain barrier, as well as promoting neurogenesis in the recovery stage following ischemic stroke. Furthermore, we identified certain matters that should be taken into account in future research, as well as proposed difficulties and opportunities in transforming TCM-derived flavonoids into medications or functional foods for the treatment or prevention of CIRI. Overall, in this review we aim to provide novel ideas for the identification of new prospective medication candidates for the therapeutic strategy against ischemic stroke.
脑缺血/再灌注损伤(CIRI)是缺血性中风预后不良的主要原因。黄酮类化合物是一大类植物多酚,在中药中含量丰富,对包括缺血性中风在内的多种疾病具有有益作用。越来越多的研究表明,源自中药的黄酮类化合物对缺血性中风后的CIRI具有缓解作用,并展现出良好的治疗潜力。在此,我们系统综述了中药黄酮类化合物的分类、代谢吸收、神经保护功效以及抗CIRI的机制。文献表明,黄酮类化合物具有潜在的药用功能,包括抑制兴奋性毒性、钙超载、氧化应激、炎症、凝血酶的细胞毒性、不同类型的程序性细胞死亡,保护血脑屏障,以及在缺血性中风后的恢复阶段促进神经发生。此外,我们确定了未来研究中应考虑的某些事项,并提出了将源自中药的黄酮类化合物转化为治疗或预防CIRI的药物或功能性食品的困难与机遇。总体而言,在本综述中,我们旨在为确定针对缺血性中风治疗策略的新的潜在药物候选物提供新思路。
