Abstract

The overdevelopment of adipose tissues, accompanied by excess lipid accumulation and energy storage, leads to adipose deposition and obesity. With the increasing incidence of obesity in recent years, obesity is becoming a major risk factor for human health, causing various relevant diseases (including hypertension, diabetes, osteoarthritis and cancers). Therefore, it is of significance to antagonize obesity to reduce the risk of obesity-related diseases. Excess lipid accumulation in adipose tissues is mediated by adipocyte hypertrophy (expansion of pre-existing adipocytes) or hyperplasia (increase of newly-formed adipocytes). It is necessary to prevent excessive accumulation of adipose tissues by controlling adipose development. Adipogenesis is exquisitely regulated by many factors in vivo and in vitro, including hormones, cytokines, gender and dietary components. The present review has concluded a comprehensive understanding of adipose development including its origin, classification, distribution, function, differentiation and molecular mechanisms underlying adipogenesis, which may provide potential therapeutic strategies for harnessing obesity without impairing adipose tissue function.

脂肪组织过度发育,伴随着脂质积累和能量储存过多,导致脂肪沉积和肥胖。近年来,随着肥胖症发病率的不断上升,肥胖症已成为人类健康的主要危险因素,引发各种相关疾病(包括高血压、糖尿病、骨关节炎和癌症)。因此,拮抗肥胖以降低肥胖相关疾病的风险具有重要意义。脂肪组织中脂质的过度积累是由脂肪细胞肥大(现有脂肪细胞的扩张)或增生(新形成的脂肪细胞的增加)介导的。通过控制脂肪发育,有必要防止脂肪组织的过度积累。脂肪生成受到体内和体外许多因素的精细调节,包括激素、细胞因子、性别和饮食成分。本综述全面总结了脂肪发育的过程,包括其起源、分类、分布、功能、分化以及脂肪生成的分子机制,这可能为在不损害脂肪组织功能的情况下控制肥胖提供潜在的治疗策略。
