Effect of thymol on oxidative stress and reelin signaling pathway in Alzheimer's disease model.

Abstract

BACKGROUND/AIM: The purpose of this study was to investigate how thymol affects cognitive functions and the levels of MDA, GSH, Aβ, ApoE, reelin, and LRP8 in an AD model induced in male Wistar albino rats with the application of D-galactose (D-gal) and aluminum chloride (AlCl).

MATERIALS AND METHODS

In this work, 3-month-old male Wistar albino rats were used. Group 1 served as the Control, Group 2 received 0.5 mL/day saline + 0.5 mL/day sunflower oil, Group 3 was administered 200 mg/kg/day AlCl + 60 mg/kg/day D-gal, Group 4 received 30 mg/kg/day thymol, and Group 5 was administered 200 mg/kg/day AlCl + 60 mg/kg/day D-gal + 30 mg/kg/day thymol. At the end of the 10-week experimental period, behavioral and memory tests were performed. GSH and MDA levels were measured in the obtained serum and brain tissue samples, while Aβ, ApoE, reelin, and LRP8 levels were measured in brain tissue samples. Statistical analyses were performed using ANOVA test in Graphpad Prism V8.3 program. A p-value <0.05 was considered significant in intergroup analyses.

RESULTS

When the novel object recognition test (NORT) results were evaluated, the Alzheimer + thymol (ALZ+TYM) group showed a significant increase in the recognition index (RI) and discrimination index (DI) compared to the Alzheimer (ALZ) group at the 24th hour. Thymol reduced working memory errors (WME), reference memory errors (RME), and maze completion time at 48, 72, and 96 hours when evaluated in terms of spatial memory in rats with Alzheimer's disease. Furthermore, Aβ and ApoE levels were increased in the ALZ group compared to the control (C), while reelin and LRP8 levels were decreased in the ALZ group compared to the C group.

CONCLUSION

The data we obtained suggest that thymol may play an effective role in cognitive processes against AD and have an anti-Alzheimer's disease effect.

背景/目的:本研究旨在探究百里香酚如何影响雄性Wistar白化大鼠应用D-半乳糖(D-gal)和氯化铝(AlCl)诱导的阿尔茨海默病(AD)模型中的认知功能以及丙二醛(MDA)、谷胱甘肽(GSH)、β-淀粉样蛋白(Aβ)、载脂蛋白E(ApoE)、reelin蛋白和低密度脂蛋白受体相关蛋白8(LRP8)的水平。

材料与方法

本研究使用3月龄雄性Wistar白化大鼠。第1组作为对照组,第2组每天接受0.5 mL生理盐水 + 0.5 mL向日葵油,第3组给予200 mg/kg/天AlCl + 60 mg/kg/天D-gal,第4组接受30 mg/kg/天百里香酚,第5组给予200 mg/kg/天AlCl + 60 mg/kg/天D-gal + 30 mg/kg/天百里香酚。在10周实验期结束时,进行行为和记忆测试。在获得的血清和脑组织样本中测量GSH和MDA水平,同时在脑组织样本中测量Aβ、ApoE、reelin蛋白和LRP8水平。使用Graphpad Prism V8.3程序中的方差分析(ANOVA)测试进行统计分析。组间分析中,p值<0.05被认为具有统计学意义。

结果

在评估新物体识别测试(NORT)结果时,阿尔茨海默病 + 百里香酚(ALZ+TYM)组在第24小时的识别指数(RI)和辨别指数(DI)相比阿尔茨海默病(ALZ)组有显著增加。当从阿尔茨海默病大鼠的空间记忆方面评估时,百里香酚在48、72和96小时减少了工作记忆错误(WME)、参考记忆错误(RME)和迷宫完成时间。此外,与对照组(C)相比,ALZ组的Aβ和ApoE水平升高,而与C组相比,ALZ组的reelin蛋白和LRP8水平降低。

结论

我们获得的数据表明,百里香酚可能在对抗AD的认知过程中发挥有效作用,并具有抗阿尔茨海默病的效果。
