Abstract

-Acetyllactosamine is a common saccharide motif found in various biologically active glycans. This motif usually works as a backbone for additional modifications and thus significantly influences glycan conformational behavior and biological activity. In this work, we have investigated the type-2 -acetyllactosamine scaffold using the complete series of its monodeoxyfluorinated analogs. These glycomimetics have been studied by molecular mechanics, quantum mechanics, X-ray crystallography, and various NMR techniques, which have provided a comprehensive and complete insight into the role of individual hydroxyl groups in the conformational behavior and lipophilicity of -acetyllactosamine.

乙酰乳糖胺是一种常见的糖基结构单元,存在于各种具有生物活性的聚糖中。这个结构单元通常作为进一步修饰的骨架,因此显著影响糖链的构象行为和生物活性。在这项工作中,我们使用全系列的单脱氧氟代类似物研究了 2 型乙酰乳糖胺支架。通过分子力学、量子力学、X 射线晶体学和各种 NMR 技术研究了这些糖模拟物,这些技术提供了对单个羟基在乙酰乳糖胺构象行为和疏水性中的作用的全面和完整的认识。
