Agmatine, 1-Amino-4-guanidinobutane, is a ubiquitous naturally occurring molecule present in low amounts in a wide variety of foodstuff. Clinical trials have demonstrated the safety of oral agmatine sulfate and have led to its development as an effective dietary ingredient for promoting resilient nerve functions. Although clearly required, the mutagenic and genotoxic effects of agmatine have not been previously reported. The present study, therefore, undertook to assess the safety profile of agmatine using currently accepted and mutagenicity and genotoxicity tests. The test item was G-Agmatine®, a proprietary brand of agmatine sulfate. Using the bacterial reverse mutation assay (Ames test), the study found that G-Agmatine® has no mutagenic effects. It had no clastogenic effects as observed by the chromosomal aberration test using Chinese Hamster lung cells. And it lacked genotoxic effects as evidenced by the lack of increased frequency of micronucleated polychromatic immature erythrocytes following oral administration in the mouse micronucleus test. Taken together with previously published data, results of the present study further support the safety of agmatine sulfate as a dietary ingredient.
胍丁胺,即1-氨基-4-胍基丁烷,是一种广泛存在于各种食物中但含量较低的天然分子。临床试验已证明口服硫酸胍丁胺的安全性,并促使其作为促进神经功能恢复的有效膳食成分得以开发。尽管显然有必要,但胍丁胺的致突变和遗传毒性作用此前尚未见报道。因此,本研究采用目前公认的致突变性和遗传毒性试验来评估胍丁胺的安全性。测试项目为G-Agmatine®,硫酸胍丁胺的一个专利品牌。通过细菌回复突变试验(艾姆斯试验),该研究发现G-Agmatine®没有致突变作用。在中国仓鼠肺细胞染色体畸变试验中未观察到其具有断裂作用。并且在小鼠微核试验中,口服给药后多染性未成熟红细胞微核频率未增加,证明其缺乏遗传毒性作用。结合此前发表的数据,本研究结果进一步支持了硫酸胍丁胺作为膳食成分的安全性。
