Abstract

Glucosinolates' species and content serve as crucial indicators for Brassicaceae plants and their products. However, endogenous enzymes in Brassicaceae can hydrolyze glucosinolates, influencing their quantitative detection. Our study utilized rapeseed, a vital Brassicaceae oil crop, to establish a novel UHPLC-MS/MS method for glucosinolate determination, incorporating a pretreatment approach with enzyme inhibition. This enabled intact glucosinolate determination at room temperature. The straightforward process involved homogenizing samples with saturated sodium chloride and methanol, followed by vortex extraction, centrifugation, and filtration. The method exhibited a strong linear relationship (R > 0.990), with LODs and LOQs ranging from 0.1 to 1.1 mg/kg and 0.3-3.7 mg/kg, respectively. Intra-day precision (7 repetitions) ranged from 3.21 to 10.47 %, while inter-day precision (3 days) ranged from 3.13 to 15.97 %.
