A bacterial strain, designated as A6, was isolated from the rhizosphere soil of a healthy muskmelon in Wenchang, Hainan Province, China. The cells of strain A6 were Gram-negative, aerobic, short rod and motile with a single polar flagellum. Strain A6 could tolerate up to 55.0 mM Al and inhibited the growth of f. sp. which is the pathogen of muskmelon . Growth occurred at 15-37 ℃ (optimum at 30 ℃), pH 4.5-8.0 (optimum pH 6.5) and with 0-3.0 % NaCl (w/v; optimum, 0.5%). Strain A6 shared the highest 16S rRNA gene sequence similarities with Sa (98.0%), followed by ATSB10 (98.0%), 5GH9-34 (97.9%), DHG59 (97.7%), DHo (97.7%) and 5GH9-11 (97.7%). Phylogenetic trees based on 16S rRNA gene and genomic sequences indicated that strain A6 belonged to the genus and formed a subclade with Sa and ATSB10. The average nucleotide identity (ANI), average amino acid identity (AAI) and digital DNA-DNA hybridization (dDDH) values between A6 and its closely related type strains were 78.8-80.8 %, 70.0-71.7 % and 20.5-22.1 %, respectively. The sole respiratory quinone was ubiquinone-8 (Q-8). The polar lipid profile consisted of diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylethanolamine (PE), two unidentified aminophospholipids (APL1-2) and three unidentified phospholipids (PL1-3). The major cellular fatty acids (≥5 %) were iso-C, C, summed feature 9 (iso-C 9 and/or C 10-methyl), iso-C, iso-C and anteiso-C. The genome size of strain A6 was 3.7 Mb with a DNA G+C content of 65.1%. Based on the phenotypic, phylogenetic, genotypic and chemotaxonomic features, strain A6 represents a novel species in the genus , for which A6 sp. nov. is proposed. The type strain is A6 (= GDMCC 1.4640 = KCTC 92542).
从中国海南省文昌市一株健康甜瓜的根际土壤中分离出一株细菌菌株,命名为A6。A6菌株的细胞为革兰氏阴性、需氧、短杆状,具有一根单极鞭毛,能运动。A6菌株能够耐受高达55.0 mM的铝,并能抑制甜瓜枯萎病菌(甜瓜的病原菌)的生长。该菌株在15-37℃(最适温度为30℃)、pH 4.5-8.0(最适pH 6.5)以及0-3.0%(w/v;最适为0.5%)NaCl条件下生长。A6菌株与嗜麦芽窄食单胞菌(98.0%)、ATSB10(98.0%)、5GH9-34(97.9%)、DHG59(97.7%)、DH0(97.7%)和5GH9-11(97.7%)的16S rRNA基因序列相似度最高。基于16S rRNA基因和基因组序列构建的系统发育树表明,A6菌株属于窄食单胞菌属,与嗜麦芽窄食单胞菌和ATSB10形成一个亚分支。A6与其密切相关的模式菌株之间的平均核苷酸同一性(ANI)、平均氨基酸同一性(AAI)和数字DNA-DNA杂交(dDDH)值分别为78.8-80.8%、70.0-71.7%和20.5-22.1%。唯一的呼吸醌为泛醌-8(Q-8)。极性脂质谱由双磷脂酰甘油(DPG)、磷脂酰甘油(PG)、磷脂酰乙醇胺(PE)、两种未鉴定的氨基磷脂(APL1-2)和三种未鉴定的磷脂(PL1-3)组成。主要的细胞脂肪酸(≥5%)为异-C15:0、C16:0、总和特征9(异-C17:0和/或C19:0环丙烷)、异-C17:1ω9c、异-C19:0和anteiso-C15:0。A6菌株的基因组大小为3.7 Mb,DNA G+C含量为65.1%。基于表型、系统发育、基因型和化学分类学特征,A6菌株代表窄食单胞菌属中的一个新物种,为此提出窄食单胞菌A6新种。模式菌株为A6(=GDMCC 1.4640 =KCTC 92542)。
