Abstract

Rhizobial isolates from Phaseolus vulgaris (common bean) grown in Heilongjiang Province were grouped into two clusters based upon the phylogenies of 16S rRNA and recA-atpD genes representing by BJ04, MJ22, MJ37, BC56, MC62 and MC63, MJ21, MJ31, respectively. The phylogenetic analysis of whole genomes further revealed that BJ04, MJ22, MJ37, BC56 and MC62 formed a cluster neighbored Rhizobium chutanense C5, while MC63, MJ21 and MJ31 formed a cluster most related with Rhizobium croatiense 13T. The whole genome average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) values between the strains BJ04 and R. chutanense C5 and between MC63 and R. croatiense 13T were lower than the species thresholds 95 % and 70 %, respectively. While the G + C content of the whole genome of the novel strains were 61.16-61.56 %, within the allowable GC content range of the Rhizobium genus (57-66 %). These results, combined with chemical classification and phenotype analysis, supported that BJ04 and MC63 represented two novel species and the names Rhizobium binxianense sp. nov. and Rhizobium mulingense sp. nov. were described for them. BJ04 (=CCTCC AB 2022367, = JCM 35885) and MC63 (=CCTCC AB 2024025, = JCM 36652) were designed as the type strains, respectively for these two novel species.

从黑龙江省种植的菜豆(普通菜豆)中分离得到的根瘤菌菌株,根据16S rRNA和recA-atpD基因的系统发育分析,分别以BJ04、MJ22、MJ37、BC56、MC62和MC63、MJ21、MJ31为代表,被分为两个簇。全基因组系统发育分析进一步表明,BJ04、MJ22、MJ37、BC56和MC62形成一个与楚坦根瘤菌C5相邻的簇,而MC63、MJ21和MJ31形成一个与克罗地亚根瘤菌13T关系最密切的簇。菌株BJ04与楚坦根瘤菌C5以及MC63与克罗地亚根瘤菌13T之间的全基因组平均核苷酸同一性(ANI)和数字DNA-DNA杂交(dDDH)值分别低于物种阈值95%和70%。而新菌株全基因组的G+C含量为61.16-61.56%,在根瘤菌属允许的GC含量范围内(57-66%)。这些结果,结合化学分类和表型分析,支持BJ04和MC63代表两个新物种,并分别为它们描述了新种宾县根瘤菌(Rhizobium binxianense sp. nov.)和穆棱根瘤菌(Rhizobium mulingense sp. nov.)。BJ04(=CCTCC AB 2022367,=JCM 35885)和MC63(=CCTCC AB 2024025,=JCM 36652)分别被指定为这两个新物种的模式菌株。
