Analyzing real-time wound healing dynamics is technically challenging. Here, we present a protocol to study epidermal cell responses in zebrafish larvae through laser-induced wounds. We describe steps for collecting embryos, mounting samples, and UV laser-assisted skin injury. We then detail procedures for live imaging of re-epithelialization and drug testing for up to 8 h and image analysis of the time-lapse footage. This protocol enables studies on cell behavior and morphological changes at injury sites and over distances.
