Abstract

This study aims to concurrently assess the blood concentrations of antiepileptic drugs including phenobarbital, carbamazepine, primidone, phenytoin in post-mortem samples through the application modified QuEChERS extraction. The serum samples were extracted and then purified using magnesium sulfate, PSA, and C18 sorbents and subsequently analyzed using HPLC/UV. One influencing parameter on the extraction process was the pH of sample. Extraction recoveries were in the range of 42-97 % for all analytes. The findings reveal that the QuEChERS method offers a higher extraction efficiency, yielding cleaner samples with greater purity and preferred choice for drug analysis in forensic toxicology. The results showed that the proposed technique has a good analytical efficiency and the accuracy of work is in the average range of 70-85 %. The equation of linear calibration curve was obtained with regression coefficient > 0.99. Detection limits were in the range of 0.21-0.38 ng/mL.•This method is inexpensive and fast for determination of antiepileptic drugs in serum•This method has high thought of sample determination.•This method is green chemistry method.

本研究旨在通过应用改良的QuEChERS提取法,同时测定死后样本中包括苯巴比妥、卡马西平、扑米酮、苯妥英在内的抗癫痫药物的血药浓度。血清样本经提取后,使用硫酸镁、PSA和C18吸附剂进行净化,随后采用HPLC/UV进行分析。提取过程中的一个影响参数是样品的pH值。所有分析物的提取回收率在42-97%范围内。研究结果表明,QuEChERS方法具有更高的提取效率,能得到纯度更高、更纯净的样品,是法医毒理学中药物分析的首选方法。结果显示,所提出的技术具有良好的分析效率,工作准确性在70-85%的平均范围内。获得了线性校准曲线方程,回归系数>0.99。检测限在0.21-0.38 ng/mL范围内。•该方法测定血清中抗癫痫药物价格低廉且快速•该方法对样品测定评价很高。•该方法是绿色化学方法。
