Abstract

This chapter explores the use of loop-mediated isothermal amplification (LAMP) for rapid and cost-effective genotyping, highlighting its advantages in field-based or resource-limited settings due to its isothermal nature. DNA is extracted from FTA samples and leaf tissue for input into a downstream LAMP assay targeting specific genes. The method's high sensitivity and specificity enables accurate detection of genotypes with simple colorimetric analysis. While challenges exist in multiplexing and non-specific amplification, this protocol highlights LAMP's simplicity and efficiency, demonstrating it is a valuable tool for plant diagnostics and breeding.
