Circular RNAs (circRNAs) are unique, abundant, and stable non-coding RNAs having regulatory roles and potential clinical significance. This study investigates the expression profiles and diagnostic efficacy of specific circRNAs in HIV-infected patients.
Blood samples from 195 HIV-infected patients (156 were viremic, 39 were non-viremic (antiretroviral therapy (ART)-suppressed) and 40 healthy controls were collected. The expression of six circRNAs in whole blood from HIV-infected patients and healthy controls was analyzed using quantitative real-time PCR. The diagnostic potential of circRNAs was assessed using receiver operating characteristic (ROC) curve analysis. Bioinformatics approaches were then used to identify competing endogenous RNA (ceRNA) networks of circRNAs with microRNAs (miRNAs) and messenger RNAs (mRNAs), their biological functions, and related pathways.
This study demonstrated that circRNAs were differentially expressed in HIV infection, highlighting that circMTO1 (viremic FC: 3.87, non-viremic FC: 3.61) and circZNF292 (viremic FC: 3.24, non-viremic FC: 3.0) may act as robust biomarkers for distinguishing HIV patients from healthy controls. The ROC curve analysis showed that for circMTO1 expression, the area under curve (AUC) value was 0.7492 (p = 0.0077), and for circZNF292 expression AUC value was 0.7231 (p = 0.0176). This study has also revealed the involvement of circMTO1 and circZNF292 in key regulatory pathways associated with immune regulation and HIV progression.
In conclusion, our findings emphasize the importance of circRNAs in understanding HIV pathogenesis and propose their clinical significance in diagnostic and therapeutic strategies.
