Adipose stem cells (ASCs) play a crucial role in maintaining adipose tissue homeostasis, but studying them in situ is technically challenging. Here, we present a protocol for the transplantation of ASCs into mouse epididymal white adipose tissue (eWAT). We describe steps for isolating and purifying ASCs, labeling them with a cell tracker, administering anesthesia, making an incision, and performing spot injections of ASCs into the eWAT of mice.
