Abstract

Oropharyngeal candidiasis (OPC) commonly occurs in immunocompromised individuals, and enhancing endogenous antimicrobial defenses with probiotics has emerged as an effective prevention strategy. Given the critical role of human β-defensin-2 (HBD-2) in combating Candida albicans infection in the oral cavity, this study aimed to identify oral commensal bacteria promoting HBD-2 production in the host. Thirty-five Streptococcus salivarius strains were screened using the HOK-16B cell line, and two were identified as significantly promoting HBD-2 secretion. These strains were further validated in a murine OPC model. The results indicated that oral intervention with S. salivarius CCFM1416 activated the IL-17/IKBζ signaling pathway in the host (IKBζ from 67.73 to 122.27 pg/mg), upregulating the expression of β-defensin (from 13.64 to 25.21 pg/mg). Consequently, C. albicans colonization and adhesion in the oral cavity were markedly reduced. Finally, a 14-day oral toxicity study demonstrated that oral administration of S. salivarius CCFM1416 is safe in vivo. In summary, the research results indicated that S. salivarius CCFM1416 can effectively prevent the occurrence of OPC and is an excellent probiotic candidate for the prevention of oral mucosal infections.

口腔念珠菌病(OPC)常见于免疫功能低下的个体,而利用益生菌增强内源性抗菌防御已成为一种有效的预防策略。鉴于人类β-防御素-2(HBD-2)在抵抗口腔白色念珠菌感染中起关键作用,本研究旨在鉴定可促进宿主产生HBD-2的口腔共生菌。使用HOK-16B细胞系筛选了35株唾液链球菌菌株,其中两株被鉴定为能显著促进HBD-2分泌。这些菌株在小鼠OPC模型中进一步得到验证。结果表明,用唾液链球菌CCFM1416进行口腔干预可激活宿主中的IL-17/IKBζ信号通路(IKBζ从67.73升至122.27 pg/mg),上调β-防御素的表达(从13.64升至25.21 pg/mg)。因此,口腔中白色念珠菌的定植和黏附明显减少。最后,一项为期14天的口服毒性研究表明,口服唾液链球菌CCFM1416在体内是安全的。总之,研究结果表明唾液链球菌CCFM1416可有效预防OPC的发生,是预防口腔黏膜感染的优秀益生菌候选菌株。
