The aim of the present study was to determine the relationship of the Thy-1 antigenic determinants to the cell surface ganglioside GM1. Exposure of either thymus or bone marrow cells to Thy-1 alloantiserum partially inhibited the binding of [125I]cholera toxin (CT) by the cells. Xenoantibodies to Thy-1 antigens failed to affect the binding of [125I]CT to mouse thymus cells. In the reverse experiments, treatment of either thymus cells or brain homogenate with CT did not alter their capacity to absorb Thy-1 allo- and xenoantibodies. It is concluded that GM1 ganglioside does not constitute a Thy-1 antigenic determinant.
本研究的目的是确定Thy-1抗原决定簇与细胞表面神经节苷脂GM1之间的关系。胸腺细胞或骨髓细胞与Thy-1同种抗血清接触会部分抑制细胞对[125I]霍乱毒素(CT)的结合。针对Thy-1抗原的异种抗体未能影响[125I]CT与小鼠胸腺细胞的结合。在反向实验中,用CT处理胸腺细胞或脑匀浆并未改变它们吸收Thy-1同种和异种抗体的能力。得出的结论是,GM1神经节苷脂不构成Thy-1抗原决定簇。
