Abstract

BACKGROUND

We evaluated possible effects of alpha-interferon (alpha-IFN) on testicular spermatogenesis and epididymal sperm quality in the nude rat.

METHODS

Nude male rats were administered subcutaneous injections of human alpha-IFN daily for 3 months. The luminal content of the cauda epididymidis was collected by micropuncture. Daily sperm production was determined by Amann's method and sperm concentrations were determined by microassay. Progressive motility was judged by evaluating the linear distance traveled by the sperm in a diluent. Serum levels of testosterone, luteinizing hormone (LH), and follicle stimulating hormone (FSH) were also measured at the end of the experiment.

RESULTS

Daily sperm production and epididymal sperm concentrations were significantly increased after administration of alpha-IFN, while progressive motility of the spermatozoa was not altered. Alpha-IFN significantly increased serum testosterone levels, while it decreased serum LH levels and left serum FSH levels unchanged.

CONCLUSION

Alpha-IFN may improve testicular spermatogenesis and increase the epididymal sperm concentration in the rat. These promising results with alpha-IFN may pave the way for a new approach to treating male infertility.

我们评估了α-干扰素(α-IFN)对裸鼠睾丸精子发生及附睾精子质量的可能影响。

方法

对裸鼠雄性大鼠每日皮下注射人α-IFN,持续3个月。通过微穿刺收集附睾尾部的管腔内容物。采用阿曼法测定每日精子生成量,通过微量测定法测定精子浓度。通过评估精子在稀释液中移动的直线距离来判断精子的前向运动能力。在实验结束时还测定了血清睾酮、黄体生成素(LH)和卵泡刺激素(FSH)水平。

结果

给予α-IFN后,每日精子生成量和附睾精子浓度显著增加,而精子的前向运动能力未改变。α-IFN显著提高血清睾酮水平,同时降低血清LH水平,血清FSH水平未变。

结论

α-IFN可能改善大鼠睾丸精子发生并增加附睾精子浓度。α-IFN的这些有前景的结果可能为治疗男性不育症开辟新途径。
