The present experiment is undertaken to study the peripheral mechanism of the inhibitory effect of intra third ventricular administration of histamine (HA, i.c.v.) on gastric acid secretion. Gastric acid was continuously washed with 37 degrees C saline by means of a perfusion pump in male adrenalectomized SD rats. Drugs were injected intravenously by a syringe pump and their effect on pentagastrin-induced [10 micrograms/(kg.h), i.v.] gastric acid secretion was measured. The results were as follows: (1) The inhibitory effect of HA on gastric acid secretion was abolished by subdiaphragmatic vagotomy. (2) The inhibitory effect of HA on gastric acid secretion could be abolished by pretreatment with atropine [0.05 mg/(kg.100 min), i.v.]. (3) Pretreatment with somatostatin antagonist, cyclo-[7-aminoheptanoyl-Phe-D-Trp-Lys-Thr (Bzl)], [2-4 micrograms/(kg.100 min), i.v.] could also block the inhibitory effect of HA on gastric acid secretion in a dose dependent manner. The above results suggest that the inhibitory effect of centrally administrated HA on gastric acid secretion is mediated by vagi, acetylcholine M receptor and somatostatin.
本实验旨在研究第三脑室内注射组胺(HA,脑室内注射)对胃酸分泌抑制作用的外周机制。在雄性肾上腺切除的SD大鼠中,通过灌注泵用37℃生理盐水持续冲洗胃酸。通过注射泵静脉注射药物,并测量其对五肽胃泌素诱导的[10微克/(千克·小时),静脉注射]胃酸分泌的影响。结果如下:(1)膈下迷走神经切断术可消除HA对胃酸分泌的抑制作用。(2)阿托品[0.05毫克/(千克·100分钟),静脉注射]预处理可消除HA对胃酸分泌的抑制作用。(3)生长抑素拮抗剂环[7-氨基庚酰-Phe-D-Trp-Lys-Thr(Bzl)] [2-4微克/(千克·100分钟),静脉注射]预处理也可剂量依赖性地阻断HA对胃酸分泌的抑制作用。上述结果表明,中枢给予的HA对胃酸分泌的抑制作用是由迷走神经、乙酰胆碱M受体和生长抑素介导的。
