To investigate the defensive effect of melatonin against oxidative stresses in ultraviolet-B (UVB) radiation induced cataract development.
Young rats received 8 kJ/m(2) UVB for 15 min. For the intervention of cataract development intraperitoneal injection of melatonin (4 mg/kg daily for 1 week) following UVB exposure was performed. Lenticular glutathione peroxidase (GSHPx), catalase (CAT), superoxide dismutase (SOD) activities and glutathione (GSH), malondialdehyde (MDA) levels were determined in UVB-melatonin, UVB, and control groups.
One week after exposure in the UVB group, lens opacities were observed and CAT, SOD, and GSHPx activities, and GSH level were lower than control and MDA level was higher than control (p < 0.05). In the UVB-melatonin group CAT and SOD activities were lower than control (p < 0.05), and the MDA level was lower than the UVB group (p < 0.05).
These results suggest that melatonin may protect against the UVB-induced cataract development by directly quenching lipid peroxides and indirectly by enhancing the production of the endogenous antioxidant GSH.
探讨褪黑素对紫外线B(UVB)辐射诱导的白内障形成过程中氧化应激的防御作用。
对年轻大鼠给予8 kJ/m(2)的UVB照射15分钟。为干预白内障形成,在UVB照射后进行腹腔注射褪黑素(每天4 mg/kg,持续1周)。测定UVB-褪黑素组、UVB组和对照组晶状体谷胱甘肽过氧化物酶(GSHPx)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性以及谷胱甘肽(GSH)、丙二醛(MDA)水平。
UVB组照射1周后,观察到晶状体混浊,CAT、SOD和GSHPx活性以及GSH水平低于对照组,MDA水平高于对照组(p < 0.05)。UVB-褪黑素组中,CAT和SOD活性低于对照组(p < 0.05),MDA水平低于UVB组(p < 0.05)。
这些结果表明,褪黑素可能通过直接淬灭脂质过氧化物以及间接增强内源性抗氧化剂GSH的产生来预防UVB诱导的白内障形成。
