To investigate the fluid shear stress induced changes of (Ca(2+))i in neutrophils in pancreatic microcirculation of experimental acute pancreatitis (AP).
Wistar rats (n = 36) were randomized into three groups. A model of AP was established by subcutaneous injection of caerulein. Low-shear 30 viscometer was used to provide steady fluid shear stress on separated neutrophils. The mean fluorescent intensity tested by flow cytometry was used as the indication of [Ca(2+)]i quantity.
Under steady shear, cytosolic [Ca(2+)]i showed biphasic changes. The shear rate changed from low to high, [(Ca(2+)]i in different groups decreased slightly and then increased gradually to a high level (P<0.05). A close correlation was observed between the cytosolic [Ca(2+)]i level and the alteration of fluid shear stress in regional microcirculation of AP.
The increase of [Ca(2+)]i is highly related to the activation of neutrophils, which contributes to neutrophil adhesion to endothelium in the early phase of AP. The effect of fluid shear stress on [Ca(2+)]i may play a crucial role in pancreatic microcirculatory failure of AP.
研究流体切应力诱导实验性急性胰腺炎(AP)胰腺微循环中中性粒细胞内钙离子(Ca(2+))i变化。
将36只Wistar大鼠随机分为三组。通过皮下注射雨蛙素建立AP模型。使用低剪切力30粘度计对分离的中性粒细胞施加稳定的流体切应力。采用流式细胞术检测平均荧光强度作为细胞内钙离子([Ca(2+)]i)含量的指标。
在稳定剪切力作用下,细胞内钙离子([Ca(2+)]i)呈现双相变化。随着剪切速率由低到高变化,不同组的细胞内钙离子([(Ca(2+)]i)先略有下降,然后逐渐升高至较高水平(P<0.05)。AP局部微循环中细胞内钙离子([Ca(2+)]i)水平与流体切应力变化密切相关。
细胞内钙离子([Ca(2+)]i)升高与中性粒细胞活化密切相关,这在AP早期有助于中性粒细胞黏附于内皮细胞。流体切应力对细胞内钙离子([Ca(2+)]i)的影响可能在AP胰腺微循环衰竭中起关键作用。
