To observe the effect of Gengnianchun Recipe (GNC) on bone mineral density (BMD), bone biomechanical parameters and serum lipid level in the bilaterally ovariectomized (OVX) rats and to explore the prophylactic and therapeutic action of GNC on ovariectomy induced osteoporosis and hyperlipidemia.
OVX SD rats, 10 - 12 months old, were divided into different groups and fed with GNC 2 g/d, GNC 1 g/d and Nilestriol 0.125 mg/week, respectively for 4 months to observe the change of BMD and bone biomechanical parameters of the lumbar vertebrae, and the serum levels of total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C), and to compare the effect of the two drugs on the morphology of the uterus.
There was marked reduction in BMD and biomechanical parameters in lumbar vertebrae (P < 0.01) and increase of serum TC and LDL-C levels (P < 0.01) in rats after OVX. GNC or Nilestriol significantly improved the decreased BMD and biomechanical parameters of the lumbar vertebrae (P < 0.05 or P < 0.01), and reduced the serum TC and LDL-C levels (P < 0.01). In the Nilestriol group, the wet weight of uterus got increased obviously (P < 0.01), the number of uterine glands increased, uterine columnar epithelium thickened, and the mitotic figures in the epithelial stroma and myointimal cells augmented. But no such effect in wet weight and morphology of uterus was found in the GNC group.
GNC could increase the BMD and biomechanical parameters of the lumbar vertebrae, reduce the serum TC and LDL-C levels, yet produce no adverse reaction in stimulating proliferation and hypertrophy of uterus.
观察更年春方(GNC)对双侧卵巢切除(OVX)大鼠骨密度(BMD)、骨生物力学参数及血脂水平的影响,探讨GNC对去卵巢所致骨质疏松症和高脂血症的防治作用。
将10 - 12月龄的OVX SD大鼠分为不同组,分别给予GNC 2 g/d、GNC 1 g/d和尼尔雌醇0.125 mg/周,连续给药4个月,观察腰椎BMD和骨生物力学参数的变化,以及血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)水平,并比较两种药物对子宫形态的影响。
OVX大鼠腰椎BMD和生物力学参数显著降低(P < 0.01),血清TC和LDL-C水平升高(P < 0.01)。GNC或尼尔雌醇能显著改善腰椎BMD和生物力学参数的降低(P < 0.05或P < 0.01),并降低血清TC和LDL-C水平(P < 0.01)。尼尔雌醇组子宫湿重明显增加(P < 0.01),子宫腺数量增多,子宫柱状上皮增厚,上皮基质和肌内膜细胞有丝分裂象增多。但GNC组子宫湿重和形态未见此效应。
GNC可提高腰椎BMD和生物力学参数,降低血清TC和LDL-C水平,且对子宫增殖和肥大无不良反应。
