Abstract

OBJECTIVE

To investigate the effects of Siwu Tang on HFCL proliferation and protein expression of bone marrow stromal cells and to discuss molecular mechanism of enriching blood functions of Siwu Tang.

METHOD

MTT, flow cytometry and proteomics were used respectively to measure the effect of Siwu Tang on HFCL proliferation, cell cycle and proteinexpression.

RESULT

Siwu Tang can promote HFCL proliferation, increase proliferative index, up-regulate 12 kinds of proteins and down-regulate 5 kinds of proteins of HFCL.

CONCLUSION

Through promoting HFCL proliferation and acting on multipleprotein targets, the blood enriching function of Siwu Tang was exerted indirectly.

摘要

目的

探讨四物汤对人脐血单个核细胞(HFCL)增殖及骨髓基质细胞蛋白表达的影响,探讨四物汤补血作用的分子机制。

方法

分别采用MTT法、流式细胞术和蛋白质组学方法检测四物汤对HFCL增殖、细胞周期及蛋白表达的影响。

结果

四物汤可促进HFCL增殖,增加增殖指数,上调HFCL的12种蛋白表达,下调5种蛋白表达。

结论

四物汤通过促进HFCL增殖并作用于多个蛋白靶点间接发挥补血作用。
