To determine butylidenephthalide in Ligusticum Chuanxiong with RP-HPLC.
The sample was extracted with methanol using sonication. The ESTD was used to quantify butylidenephthalide. HPLC separation was carried out in a Hypersil ODS columm (4.6 mm x 150 mm, 5 microm) , eluted at 1 mL x min(-1) with methanol-5% isopropyl alcohol (60: 40) at 25 degrees C. The detection wavelength was 230 nm.
The linear range was 0.07-0.7 microg for butylidenephthalide. The average recovery was 95.3%, and RSD was 2.3% (n =6).
This method was simple and could be used to determine butylidenephthalide with satisfactory accuracy and reproducibility.
采用反相高效液相色谱法(RP-HPLC)测定川芎中丁基苯酞的含量。
样品用甲醇超声提取。采用外标法对丁基苯酞进行定量。在Hypersil ODS柱(4.6 mm×150 mm,5μm)上进行HPLC分离,于25℃以1 mL·min⁻¹的流速用甲醇-5%异丙醇(60∶40)洗脱,检测波长为230 nm。
丁基苯酞的线性范围为0.07 - 0.7μg。平均回收率为95.3%,相对标准偏差(RSD)为2.3%(n = 6)。
该方法简便,可用于测定丁基苯酞,准确性和重现性良好。
