To analyze the toxic characteristic of microcystis in a life water source.
Microcystis in Huayuankou conservation pool and Xiliu lake were isolated by 96-microbiological assay and supreme dilution methods. Phycocyanin intergenic spacer region (PC-IGS) and microcystin synthetase gene B (McyB) were detected by whole-cell PCR method, And toxicities of microcystis were identified by solid pole of extraction (SPE) and high performance liquid chromatography (HPLC) method.
The microcystis of the water source were cyanobacteria which could contain PC-IGS and McyB expressing positively, Comparition with reported meyB of Microcystis aeruginosa in Genbank, the homology of gene sequence was 99%. The main toxic isomers of the three microcystis were MC-LR, whose proportions of the whole toxin were 97.9%, 98.6% and 99.3%, respectively.
The microcystis of the main life water source maybe poisonous and the main isomer maybe MC-LR.
分析某生活饮用水水源中微囊藻的毒性特征。
采用96孔微生物检测法和最高稀释法对花园口蓄水池和西流湖中的微囊藻进行分离。采用全细胞PCR法检测藻蓝蛋白基因间隔区(PC-IGS)和微囊藻毒素合成酶基因B(McyB),并通过固相萃取(SPE)和高效液相色谱(HPLC)法鉴定微囊藻的毒性。
该水源中的微囊藻为蓝藻,可检测到PC-IGS和McyB呈阳性表达,与Genbank中已报道的铜绿微囊藻的meyB相比,基因序列同源性为99%。三种微囊藻的主要毒性异构体均为MC-LR,其在总毒素中的比例分别为97.9%、98.6%和99.3%。
该主要生活饮用水水源中的微囊藻可能有毒,主要异构体可能为MC-LR。
