To study the effect of basic fibroblast growth factor (bFGF) on the gene expression of decorin by periodontal ligament fibroblasts (PLFs) in culture, and discuss the effect of bFGF in periodontal regeneration.
Human PLFs were cultured and stimulated by exogenous bFGF. Gene expression of decorin was assessed by semi-quantitive RT-PCR.
The mRNA expression of decorin was suppressed by bFGF and the effect was dose-dependent. When the dose of bFGF increased, the inhibitive effect decreased.
Decorin has many biological effects. The inhibitive effect may be one of important factors which participate in the healing process of periodontitis, and provide partly theoretical basis of bFGF in periodontal regeneration.
研究碱性成纤维细胞生长因子(bFGF)对体外培养的牙周膜成纤维细胞(PLFs)中核心蛋白聚糖基因表达的影响,并探讨bFGF在牙周组织再生中的作用。
培养人牙周膜成纤维细胞并给予外源性bFGF刺激。采用半定量逆转录聚合酶链反应(RT-PCR)检测核心蛋白聚糖的基因表达。
bFGF抑制核心蛋白聚糖的mRNA表达,且呈剂量依赖性。随着bFGF剂量增加,抑制作用减弱。
核心蛋白聚糖具有多种生物学效应。其抑制作用可能是参与牙周炎愈合过程的重要因素之一,为bFGF在牙周组织再生中的应用提供了部分理论依据。
