Abstract

OBJECTIVE

To compare alpha-glycosidase inhibitor microamount screening models which are established by two different resources alpha-glycosidase to screen the Chinese herbal medicines which have great alpha-glycosidase inhibition.

METHODS

Comparing the activities of two different resources alpha-glycosidase with glucose oxidase method, then establishing optimal reaction conditions. Extracting the water soluble compositions of Chinese gallnut, paeoniae radix, Glycyrrhiza, rhubarb, fructus by boiling ,then using the alpha-glycosidase inhibition model to check and compare their alpha-glycosidase inhibitions.

RESULTS

Five Chinese herbal medicines all had alpha-glycosidase inhibition, and the greatest was Chinese gallnut.

CONCLUSION

The activities of two resources alpha-glycosidase are certainly different, but they do not have significant influence on findings of screening alpha-glycosidase inhibitors in vitro. Water soluble compositions of five Chinese herbal medicines have alpha-glycosidase inhibition.

目的

比较由两种不同来源的α-糖苷酶建立的α-糖苷酶微量筛选模型,以筛选出具有较强α-糖苷酶抑制作用的中药。

方法

用葡萄糖氧化酶法比较两种不同来源α-糖苷酶的活性,进而建立最佳反应条件。采用水煮法提取五倍子、白芍、甘草、大黄、果实的水溶性成分,然后用α-糖苷酶抑制模型检测并比较它们对α-糖苷酶的抑制作用。

结果

五种中药均有α-糖苷酶抑制作用,其中五倍子的抑制作用最强。

结论

两种来源的α-糖苷酶活性确实存在差异,但对体外筛选α-糖苷酶抑制剂的结果没有显著影响。五种中药的水溶性成分具有α-糖苷酶抑制作用。
