To investigate the effects of Liuweidihuang Decoction on the proliferation and melanin synthesis of cultured human melanocytes in vitro.
MTT assay was used to assess the proliferation of cultured human melanocytes after treatment with Liuweidihuang Decoction, and NaOH assay was employed to determine melanin synthesis.
Liuweidihuang Decoction at the concentration of 5%-15% did not affect the proliferation of the melanocytes (P>0.05), and the decoction at 20%-30% significantly inhibited the cell proliferation, especially at the optimal concentration of 30% (P<0.01). The decoction promoted melanin synthesis in the melanocytes at the concentration of 10%-15% with the optimal concentration of 15% (P<0.01) but failed to produce such an effect at 20%-30%.
Liuweidihuang Decoction exert a two-way regulation on the proliferation and melanin synthesis of cultured human melanocytes in vitro.
探讨六味地黄汤对体外培养的人黑素细胞增殖及黑素合成的影响。
采用MTT法检测六味地黄汤作用后培养的人黑素细胞的增殖情况,采用NaOH法测定黑素合成。
5%-15%浓度的六味地黄汤对黑素细胞增殖无影响(P>0.05),20%-30%浓度的六味地黄汤显著抑制细胞增殖,其中30%浓度时抑制作用最为明显(P<0.01)。10%-15%浓度的六味地黄汤可促进黑素细胞合成黑素,其中15%浓度时促进作用最为明显(P<0.01),而20%-30%浓度时则无此作用。
六味地黄汤对体外培养的人黑素细胞的增殖及黑素合成具有双向调节作用。
