Propolis is a beehive product and its immunomodulatory action has been well documented; however, little is known concerning its activity on the immune system of stressed mice. This work investigated a possible role of propolis against the immunosuppressive effects induced by stress in mice, assessing the pro-inflammatory cytokine (IL-1beta and IL-6) production and Toll-like receptor (TLR-2 and TLR-4) expression by spleen cells.
BALB/c mice were divided into 3 groups: G1 was considered control; G2 was submitted to restraint stress for 3 days, and G3 was treated with propolis and immediately submitted to stress. After sacrifice, spleens were removed and TLR-2 and TLR-4 gene expression was analyzed, as well as the pro-inflammatory cytokine production. Serum corticosterone levels were determined by radioimmunoassay as a stress indicator.
Stressed mice, treated or not with propolis, produced higher corticosterone levels, whereas IL-1beta and IL-6 production was inhibited. TLR-2 and TLR-4 expression was inhibited in stressed mice, while propolis exerted an immunorestorative role in TLR-4 expression. The immunosuppressive effects on IL-1beta and IL-6 production and on TLR expression by stressed mice might have occurred due to a higher corticosterone production during stress.
Propolis treatment did not antagonize the inhibitory effects on pro-inflammatory cytokine production, however it restored at least partially TLR2 mRNA expression and counteracted the inhibition on TLR-4 expression in stressed animals, contributing to the recognition of microorganisms during stressful conditions.

将 BALB/c 小鼠分为 3 组:G1 为对照组;G2 接受 3 天的束缚应激,G3 用蜂胶处理并立即进行应激。处死后取出脾脏,分析 TLR-2 和 TLR-4 基因表达以及促炎细胞因子的产生。用放射免疫法测定血清皮质酮水平作为应激指标。
应激小鼠,无论是否用蜂胶处理,皮质酮水平均升高,而 IL-1β和 IL-6 的产生受到抑制。TLR-2 和 TLR-4 的表达在应激小鼠中受到抑制,而蜂胶在 TLR-4 的表达中发挥了免疫修复作用。应激小鼠对 IL-1β和 IL-6 产生和 TLR 表达的免疫抑制作用可能是由于应激期间皮质酮产生增加所致。
蜂胶处理不能拮抗对促炎细胞因子产生的抑制作用,但它至少部分恢复了应激动物 TLR2 mRNA 的表达,并抑制了 TLR-4 的表达,有助于在应激条件下识别微生物。
