Abstract

OBJECTIVE

To explore the mechanisms of Xiaotan Sanjie Decoction (XTSJD), a compound traditional Chinese herbal medicine, in inhibiting the tumor growth and preventing recurrence by testing the protein expressions of interleukin-8 (IL-8) and its receptors chemokine receptor 1 (CXCR1) and chemokine receptor 2 (CXCR2) in gastric tumor xenografts and gastric tissue adjacent to the tumor in mice.

METHODS

Fifty Kunming mice were randomly divided into normal group, normal saline (NS) group, Heat-clearing and Detoxicating Decoction (HCDD) group, tegafur (FT-207) group and XTSJD group. Except for mice in the normal group, S180 tumor block was transplanted into the gastric walls of the mice, and the mice were administered with corresponding medicine for 3 weeks. Weight of tumor xenografts was measured and tumor inhibition rate was calculated. IL-8 protein expression was tested by enzyme-linked immunosorbent assay. Expressions of CXCR1 and CXCR2 were tested by immunohistochemical method.

RESULTS

The protein expressions of IL-8 and its receptors in tumor xenografts and gastric tissue adjacent to the tumor were markedly higher than those in the gastric tissue in normal mice (P<0.01); compared with HCDD and FT-207, XTSJD could significantly decrease the IL-8 protein expression in tumor xenografts and gastric tissue adjacent to the tumor (P<0.05); compared with FT-207, XTSJD could significantly decrease the CXCR1 protein expression in tumor xenografts (P<0.01), and XTSJD could also significantly decrease the CXCR1 protein expression in gastric tissue adjacent to the tumor as compared with HCDD and FT-207 (P<0.01); compared with HCDD and FT-207, XTSJD could significantly decrease the CXCR2 protein expression in tumor xenografts (P<0.01), and there was no significant difference among the three drug-treated groups in CXCR2 protein expression in gastric tissue adjacent to the tumor (P>0.05).

CONCLUSION

XTSJD can decrease the protein expressions of IL-8 and its receptors in tumor xenografts and gastric tissue adjacent to the tumor. It may be one of the mechanisms of XTSJD in inhibiting the tumor growth and preventing recurrence.

目的

通过检测白细胞介素-8(IL-8)及其受体趋化因子受体1(CXCR1)和趋化因子受体2(CXCR2)在小鼠胃肿瘤异种移植瘤及肿瘤旁胃组织中的蛋白表达,探讨中药复方消痰散结方抑制肿瘤生长及预防复发的机制。

方法

将50只昆明小鼠随机分为正常组、生理盐水(NS)组、清热解毒方(HCDD)组、替加氟(FT-207)组和消痰散结方(XTSJD)组。除正常组小鼠外,其余小鼠胃壁移植S180肿瘤块,并给予相应药物治疗3周。测量肿瘤异种移植瘤重量并计算抑瘤率。采用酶联免疫吸附测定法检测IL-8蛋白表达。采用免疫组织化学方法检测CXCR1和CXCR2的表达。

结果

肿瘤异种移植瘤及肿瘤旁胃组织中IL-8及其受体的蛋白表达明显高于正常小鼠胃组织(P<0.01);与HCDD组和FT-207组相比,XTSJD能显著降低肿瘤异种移植瘤及肿瘤旁胃组织中IL-8蛋白表达(P<0.05);与FT-207组相比,XTSJD能显著降低肿瘤异种移植瘤中CXCR1蛋白表达(P<0.01),且与HCDD组和FT-207组相比,XTSJD也能显著降低肿瘤旁胃组织中CXCR1蛋白表达(P<0.01);与HCDD组和FT-207组相比,XTSJD能显著降低肿瘤异种移植瘤中CXCR2蛋白表达(P<0.01),而肿瘤旁胃组织中CXCR2蛋白表达在三个药物治疗组之间差异无统计学意义(P>0.05)。

结论

消痰散结方能降低肿瘤异种移植瘤及肿瘤旁胃组织中IL-8及其受体的蛋白表达。这可能是消痰散结方抑制肿瘤生长及预防复发的机制之一。
