To explore the expression of heme oxygenase-1 (HO-1) in the peripheral blood mononuclear cell (PBMC) and relationship to ventilatory function in asthmatic patients.
Eighteen asthmatic patients and eighteen healthy subjects were selected. HO-1 protein levels in PBMC were measured by immunohistochemical staining and PBMC HO-1 mRNA were determined by reverse transcription-polymerase chain reaction (RT-PCR), blood carbon monoxide Hb (COHb) percent value, serum total IgE concentration and pulmonary ventilatory function were observed in asthmatic patients and healthy subjects.
The percentage of cells in immunohistochemical staining positive staining of HO-1 were significantly higher in asthmatic patients (41.7 +/- 7.44%) compared with that of healthy subjects (10.5 +/- 4.36%, P < 0.01), the optical densities of PBMC HO-1 mRNA were higher in asthmatic patients (26.05 +/- 4.14) compared with that of healthy subjects (10.82 +/- 4.26, P < 0.01). The relation analysis showed PBMC HO-1 protein levels had significantly negative relation with FEV1, PEFR, MEFR50%, respectively (r = -0.89, -0.56, -0.51, P < 0.01, respectively) and positive relation with COHb percent value, serum total I gE concentration (r = 0.80, 0.48, P < 0.05, respectively), and PBMC HO-1 mRNA levels had significantly negative relation with FEV1, PEFR, MEFR50%, respectively (r = -0.89, -0.65, -0.67, P < 0.05, respectively) and positive relation with COHb percent value, serum total IgE concentration (r = 0.85, 0.62, P < 0.01, respectively).
The expression of PBMC HO-1 protein and mRNA are increased significantly in asthmatic patients, HO-1 may play a significant role in the pathogenesis of asthma. The expression of HO-1 has relation with severity of asthma.
探讨血红素加氧酶-1(HO-1)在外周血单个核细胞(PBMC)中的表达及其与哮喘患者通气功能的关系。
选取18例哮喘患者和18例健康对照者。采用免疫组织化学染色法检测PBMC中HO-1蛋白水平,逆转录聚合酶链反应(RT-PCR)法测定PBMC中HO-1 mRNA水平,观察哮喘患者和健康对照者的血中一氧化碳血红蛋白(COHb)百分比值、血清总IgE浓度及肺通气功能。
哮喘患者HO-1免疫组织化学染色阳性细胞百分比(41.7±7.44%)显著高于健康对照者(10.5±4.36%,P<0.01),哮喘患者PBMC中HO-1 mRNA光密度值(26.05±4.14)高于健康对照者(10.82±4.26,P<0.01)。相关性分析显示,PBMC中HO-1蛋白水平分别与第一秒用力呼气容积(FEV1)、呼气峰流速(PEFR)、50%最大呼气中期流速(MEFR50%)呈显著负相关(r分别为-0.89、-0.56、-0.51,P均<0.01),与COHb百分比值、血清总IgE浓度呈正相关(r分别为0.80、0.48,P均<0.05);PBMC中HO-1 mRNA水平分别与FEV1、PEFR、MEFR50%呈显著负相关(r分别为-0.89、-0.65、-0.67,P均<0.05),与COHb百分比值、血清总IgE浓度呈正相关(r分别为0.85、0.62,P均<0.01)。
哮喘患者PBMC中HO-1蛋白和mRNA表达显著增加,HO-1可能在哮喘发病机制中起重要作用,其表达与哮喘严重程度有关。
