To study optic nerve lesions, changes in blood flow to the optic nerve, and permeability of micro blood vessels and histopathology in diabetic rats.
Male Wistar rats (n=20) were randomly divided into control and diabetic groups. The diabetic model was prepared by a single injection of streptozotocin (50mg/kg) into the caudal vein. Three months later, laser Doppler perfusion imaging was used to observe the changes in blood flow to the optic nerve. Each rat was injected with 15g/L Evans blue (5µL/g). The permeability of microvessels in diabetic optic nerves was measured by spectrophotometry. Optic nerves were observed by light and transmission electron microscopy.
Diabetic rats had atrophic optic nerve fibers with neurite swelling, loss of myelin, and a greater-than-normal proliferation of astrocytes, occurring within 3 months of induction of diabetes. Blood flow to the optic nerve was lower in diabetic rats than in controls. Microvessel permeability in diabetic rats increased 2.03-fold compared to controls.
Diabetic rats develop significant pathological changes in the optic nerve, reduced blood flow to the optic nerve and increase microvessel permeability.
研究糖尿病大鼠的视神经病变、视神经血流变化、微血管通透性及组织病理学变化。
将20只雄性Wistar大鼠随机分为对照组和糖尿病组。通过尾静脉单次注射链脲佐菌素(50mg/kg)制备糖尿病模型。3个月后,采用激光多普勒灌注成像观察视神经血流变化。每只大鼠注射15g/L伊文思蓝(5µL/g)。用分光光度法测量糖尿病大鼠视神经微血管的通透性。通过光镜和透射电镜观察视神经。
糖尿病大鼠在糖尿病诱导后3个月内出现视神经纤维萎缩、神经突肿胀、髓鞘丢失以及星形胶质细胞异常增殖。糖尿病大鼠的视神经血流低于对照组。糖尿病大鼠的微血管通透性比对照组增加了2.03倍。
糖尿病大鼠视神经出现明显的病理变化,视神经血流减少,微血管通透性增加。
