采用大脑中动脉闭塞大鼠模型,运用脑微透析和高效液相色谱荧光检测技术,收集大鼠纹状体细胞外液中氨基酸神经递质,观察3'-甲氧基葛根素处理前后四种氨基酸的变化。
3'-甲氧基葛根素降低了兴奋性氨基酸(EAA)天冬氨酸(Asp)和谷氨酸(Glu)的浓度,而抑制性氨基酸牛磺酸(Tau)和γ-氨基丁酸(GABA)显著降低。
3'-甲氧基葛根素减轻缺血诱导的脑EAA毒性,对抗EAA神经毒性,调节脑内神经递质氨基酸含量,改善兴奋性和抑制性氨基酸平衡,是其改善和保护大鼠急性脑梗死脑核团的重要机制之一。
