Abstract

OBJECTIVE

To explore the relationship between IL-1β, IL-1Ra gene polymorphism and the occurrence of polycystic ovary syndrome (PCOS) infertility.

METHODS

A total of 59 PCOS infertility cases visiting the reproductive center of our hospital from Mar. 2010 to Mar. 2012 and 56 healthy women were selected. ELISA method was used for the detection of IL-1β, IL-1Ra levels, and the levels of serum supersensitivity C reaction protein (US-CRP), insulin (FINS), follicule-stimulating hormone (FSH) and fasting blood-glucose (FBG) were detected. PCR analysis technology was adopted to detect the gene polymorphism of the 511 site of IL-1β and the second introne of IL-1Ra.

RESULTS

The levels of IL-1β, IL-1Ra, US-CRP, FINS and FBG in blood serum of patients in PCOS group were significantly higher than those in control group (P<0.05 or P<0.01). The level of FSH in PCOS group was significantly lower than that in control group (P<0.05). The genotypic frequency of T/T, the 511 site of IL-1β in PCOS group was 42.37%, significantly higher than 12.50% in control group (P<0.01). The frequency of T allele was also significantly higher than that in control group (P<0.01). The genotypic frequency of I/V, the second introne of IL-1Ra in PCOS group was 20.34%, signiciantly higher than 3.57% in control group (P<0.05). The frequency of V allele in PCOS group was significantly higher than that in control group (P<0.05).

CONCLUSIONS

T allele of the 511 site of IL-1β gene and V allele of the second introne of IL-1Ra gene might be the genetic basis of the rising of IL-1β, IL-1Ra and US-CRP levels in blood serum of PCOS patients, and are associated with the infertility occurrence of PCOS patients.

目的

探讨白细胞介素-1β(IL-1β)、白细胞介素-1 受体拮抗剂(IL-1Ra)基因多态性与多囊卵巢综合征(PCOS)不孕的关系。

方法

选择 2010 年 3 月至 2012 年 3 月在我院生殖中心就诊的 59 例 PCOS 不孕患者和 56 例健康女性,采用 ELISA 法检测血清中 IL-1β、IL-1Ra 水平,检测血清超敏 C 反应蛋白(US-CRP)、胰岛素(FINS)、卵泡刺激素(FSH)和空腹血糖(FBG)水平。采用 PCR 分析技术检测 IL-1β 第 511 位和 IL-1Ra 第二内含子的基因多态性。

结果

PCOS 组患者血清中 IL-1β、IL-1Ra、US-CRP、FINS 和 FBG 水平明显高于对照组(P<0.05 或 P<0.01)。PCOS 组 FSH 水平明显低于对照组(P<0.05)。IL-1β 第 511 位 TT 基因型频率为 42.37%,明显高于对照组的 12.50%(P<0.01)。T 等位基因频率也明显高于对照组(P<0.01)。IL-1Ra 第二内含子 II 基因型频率为 20.34%,明显高于对照组的 3.57%(P<0.05)。PCOS 组 V 等位基因频率明显高于对照组(P<0.05)。

结论

IL-1β 基因第 511 位 T 等位基因和 IL-1Ra 基因第二内含子 V 等位基因可能是 PCOS 患者血清中 IL-1β、IL-1Ra 和 US-CRP 水平升高的遗传基础,与 PCOS 患者不孕的发生有关。
