Asthma is the most common disease in children, and its pathogenesis is highly complicated.
CD4+ cells and CD4+ IL-17+ cells were analysed by flow cytometry, and the ratio of CD4+ cells to the peripheral blood mononuclear cells (PBMCs) and CD4+ IL-17+ cells to PMBCs were calculated from the control group, acute group, and moderate group. The levels of protein expression of IL-6, IL-17, and IL-21 were detected by ELISA in the plasma and culture supernatants of PBMCs of the three groups. The correlations between IL-7 and IL-6, IL-7 and IL-21 were analysed in culture supernatants of PBMCs of the three groups.
The mean ratio of CD4+ IL-17+ cells/peripheral blood mononuclear cells (PBMCs) was 4.32% in the acute group, which was higher than in the control group and moderate group. The levels of IL-6 and IL-17 were elevated, and the levels of IL-21 were decreased in the acute group. The levels of IL-17 and IL-6 were positively correlated (r = 0.182, p = 0.03), and the levels of IL-17 and IL-21 were negatively correlated (r = -0.834, p = 0.02).
The results suggest that the levels of IL-17 and IL-6 are related to the severity of asthma. Furthermore, IL-17 may play a role in the development of childhood asthma.
哮喘是儿童最常见的疾病,其发病机制高度复杂。
采用流式细胞术分析CD4+细胞和CD4+ IL-17+细胞,并计算对照组、急性组和中度组中CD4+细胞与外周血单个核细胞(PBMCs)的比例以及CD4+ IL-17+细胞与PMBCs的比例。采用酶联免疫吸附测定法(ELISA)检测三组PBMCs血浆和培养上清液中IL-6、IL-17和IL-21的蛋白表达水平。分析三组PBMCs培养上清液中IL-7与IL-6、IL-7与IL-21之间的相关性。
急性组CD4+ IL-17+细胞/外周血单个核细胞(PBMCs)的平均比例为4.32%,高于对照组和中度组。急性组IL-6和IL-17水平升高,IL-21水平降低。IL-17与IL-6水平呈正相关(r = 0.182,p = 0.03),IL-17与IL-21水平呈负相关(r = -0.834,p = 0.02)。
结果表明,IL-17和IL-6水平与哮喘严重程度相关。此外,IL-17可能在儿童哮喘的发生发展中起作用。
