Abstract

OBJECTIVE

To determine the antitumor activity of Radix tetrastigmae flavonoids and their inhibitory effect on regulatory T cells (Tregs) in mice.

MATERIALS AND METHODS

Total flavonoids were isolated from Radix tetrastigmae, the root of Tetrastigma hemsleyanum Diels et Gilg, and administered to C57BL/6 mice by oral gavage after inoculation with Lewis lung carcinoma (LLC) cells. The effects of total flavonoids on tumor growth in vivo were examined. Flow cytometry was used to study the effects on Tregs, and enzyme-linked immunosorbent assay was used to analyze the changes in the serum levels of transforming growth factor β, prostaglandin E2, and cyclooxygenase 2 after tumor inoculation and flavonoid administration.

RESULTS

Total flavonoids from T. hemsleyanum Diels et Gilg significantly inhibited tumor growth in C57BL/6 mice inoculated with LLCs. These flavonoids dramatically suppressed regulatory T-cell development in tumor-bearing mice. Further studies revealed that total flavonoids significantly decreased the serum levels of transforming growth factor β, prostaglandin E2, and cyclooxygenase 2 in tumor-bearing mice, which may be responsible for the inhibition of Tregs.

CONCLUSION

The antitumor activity of total flavonoids from T. hemsleyanum Diels et Gilg is associated with the inhibition of Tregs in a mouse tumor model. Total flavonoids from T. hemsleyanum Diels et Gilg may be used as antitumor agents in cancer prevention and treatment.

目的

确定三叶崖爬藤黄酮的抗肿瘤活性及其对小鼠调节性T细胞(Tregs)的抑制作用。

材料与方法

从三叶崖爬藤(Tetrastigma hemsleyanum Diels et Gilg)的根中分离出总黄酮,在接种Lewis肺癌(LLC)细胞后通过口服灌胃给予C57BL/6小鼠。检测总黄酮对体内肿瘤生长的影响。采用流式细胞术研究其对Tregs的影响,并用酶联免疫吸附测定法分析接种肿瘤和给予黄酮后血清中转化生长因子β、前列腺素E2和环氧合酶2水平的变化。

结果

三叶崖爬藤总黄酮显著抑制接种LLC的C57BL/6小鼠的肿瘤生长。这些黄酮显著抑制荷瘤小鼠调节性T细胞的发育。进一步研究表明,总黄酮显著降低荷瘤小鼠血清中转化生长因子β、前列腺素E2和环氧合酶2的水平,这可能是其抑制Tregs的原因。

结论

三叶崖爬藤总黄酮的抗肿瘤活性与在小鼠肿瘤模型中抑制Tregs有关。三叶崖爬藤总黄酮可作为癌症预防和治疗中的抗肿瘤药物。
