The purpose of vitro study was to assessment of the bacterial contamination on daily twice-used toothbrushes in different conditions after 1-month and 3 months.
Forty children aged between 6 and 12 years were selected for this study. Toothbrushes and toothpaste were distributed among those children. Among those children, 20 children brushed their teeth for 1-month and 20 brushed their teeth for 3 months twice a day. Among those, 10 were kept in same brush box with a family member and 10 were kept in separate brush box after the use. Toothbrush from every child was recollected to investigate the contamination of microorganisms. Head of the toothbrushes transferred to a tube containing 10 ml of tryptone soy broth. This was followed by vortex mixing for 1 min to dislodge suspected adherent bacteria. The bacterial suspension was serially diluted to obtain dilution factors of up to 10(-3). 1 ml each of the dilution factors was obtained using a sterile pipette and plated on plate count agar. Petri dishes containing agar media were incubated and examined using a compound microscope.
There was high mean difference between 1-month and 3 months, twice a day used toothbrush those kept with family members and those kept separate.
Toothbrush should keep in a separate box. Moreover, toothbrush should to be change after 3-4 weeks.
本体外研究的目的是评估在不同条件下每日使用两次的牙刷在1个月和3个月后细菌污染情况。
本研究选取了40名6至12岁的儿童。向这些儿童分发了牙刷和牙膏。在这些儿童中,20名儿童每天刷牙两次,为期1个月,20名儿童每天刷牙两次,为期3个月。其中,10名儿童与家庭成员共用一个牙刷盒,10名儿童使用后将牙刷放在单独的牙刷盒中。收集每个儿童的牙刷以调查微生物污染情况。将牙刷头部转移至装有10毫升胰蛋白胨大豆肉汤的试管中。随后进行涡旋混合1分钟,以去除疑似附着的细菌。将细菌悬液进行系列稀释以获得高达10(-3)的稀释倍数。使用无菌移液管分别吸取1毫升各稀释倍数的菌液,接种于平板计数琼脂培养基上。将含有琼脂培养基的培养皿进行培养,并使用复式显微镜进行检查。

牙刷应放在单独的盒子里。此外,牙刷应在3至4周后更换。
