oncogene is a member of the polycomb-group gene family and a transcriptional repressor. Overexpression of BMI-1 has been identified in various human cancer tissues and is known to be involved in cancer cell proliferation, cell invasion, distant metastasis, chemosensitivity and patient survival. Accumulating evidence has revealed that BMI-1 is also involved in the regulation of self-renewal, differentiation and tumor initiation of cancer stem cells (CSCs). However, the molecular mechanisms underlying these biological processes remain unclear. The present review summarized the function of BMI-1 in different human cancer types and CSCs, and discussed the signaling pathways in which BMI-1 is potentially involved. In conclusion, BMI-1 may represent a promising target for the prevention and therapy of various cancer types.
BMI-1是多梳蛋白家族的成员之一,是一种转录抑制因子。BMI-1在多种人类癌症组织中均有过表达,已知其参与癌细胞增殖、细胞侵袭、远处转移、化疗敏感性及患者生存。越来越多的证据表明,BMI-1还参与癌症干细胞(CSC)的自我更新、分化及肿瘤起始的调控。然而,这些生物学过程背后的分子机制仍不清楚。本综述总结了BMI-1在不同人类癌症类型及CSC中的功能,并讨论了BMI-1可能参与的信号通路。总之,BMI-1可能是预防和治疗多种癌症类型的一个有前景的靶点。
