Abstract

A novel haloalkalitolerant, rod-shaped bacterium, designated strain YIM 4-4, was isolated from the surface water of the Dugerno lake, a haloalkaline lake in Inner Mongolia. The taxonomy of strain YIM 4-4 was investigated by a polyphasic approach. Strain YIM 4-4 was Gram-stain-negative, strictly aerobic, non-motile and formed red colonies. Optimal growth conditions were 28 °C, pH 8.0-11.0 and 0.5-2 % NaCl. The major respiratory quinone was menaquinone-7 (MK-7). The polar lipid profile was composed predominantly of phosphatidylethanolamine, six unidentified polar lipids, one phospholipid and one aminolipid. The predominant cellular fatty acids (>5 %) were iso-C, iso-CI/anteiso-CB, iso-CG, iso-C 3-OH, Cω7/Cω6 and iso-C. The genomic DNA G+C content was 43.0 mol%. 16S rRNA gene sequence analysis indicated that the members of the genera , , and of the family (phylum ) were the most closely related, with 16S rRNA gene sequence similarities ranging from 93.6 to 94.2 %. Other members of the family showed sequence similarities < 93.0 %. On the basis of phenotypic, chemotaxonomic and phylogenetic properties, strain YIM 4-4 represents a novel species of a new genus, for which the name gen. nov., sp. nov. is proposed. The type strain is YIM 4-4 ( = CCTCC AB 2012966 = DSM 27929).

从内蒙古一个盐碱湖——杜格诺湖的地表水分离出一株新型耐盐碱、杆状细菌,命名为菌株YIM 4-4。采用多相分类法对菌株YIM 4-4进行分类研究。菌株YIM 4-4革兰氏染色阴性,严格好氧,无运动性,形成红色菌落。最适生长条件为28 °C、pH 8.0 - 11.0和0.5 - 2 % NaCl。主要呼吸醌为甲基萘醌-7(MK-7)。极性脂质谱主要由磷脂酰乙醇胺、六种未鉴定的极性脂质、一种磷脂和一种氨基脂质组成。主要细胞脂肪酸(>5 %)为异-C、异-CI/anteiso-CB、异-CG、异-C 3-OH、Cω7/Cω6和异-C。基因组DNA G+C含量为43.0 mol%。16S rRNA基因序列分析表明,(门)中、、和属的成员亲缘关系最密切,16S rRNA基因序列相似性范围为93.6%至94.2 %。科的其他成员序列相似性<93.0 %。基于表型、化学分类和系统发育特性,菌株YIM 4-4代表一个新属的一个新物种,为此提出新属名,新种名。模式菌株为YIM 4-4(=CCTCC AB 2012966 = DSM 27929)。
