Abstract

Enzyme-assisted extraction process of the water-soluble Malva sylvestris polysaccharides (MSPs) was optimized using response surface methodology (RSM). The highest yield (10.40%) of MSPs was achieved at 5.64% cellulase, 55.65°C temperature, 3.4h time, and 5.22 pH. Three homogeneous polysaccharide fractions (MSP-1, MSP-2, MSP-3) were purified by DEAE-cellulose and Sephadex G-100 chromatography, which were composed of galactose, glucuronic acid, arabinose, rhamnose and mannose in different molar ratios with molecular weight range of 2.6×10-8.8×10Da. The fractions could significantly increase antioxidant, antitumor and antimicrobial activities in a dose-dependent pattern. MSP-2 revealed stronger antioxidant activities than MSP-1 and MSP-3, including reducing power and scavenging activity of DPPH and OH radicals. The antiproliferative activity of MSP-2 (1.0mg/mL) on the growth of A549 and HepG2 cells was 45.1% and 53.2%, respectively. The Gram-positive bacteria (Bacillus cereus PTCC 1015 and Staphylococcus aureus PTCC 1112) compared with Gram-negative ones (Escherichia coli PTCC 1763 and Salmonella typhimurium PTCC 1709) showed less sensitivity against the various MSPs (3-15mg/mL).

采用响应面法(RSM)对锦葵水溶性多糖(MSPs)的酶辅助提取工艺进行了优化。在纤维素酶用量为 5.64%、温度为 55.65°C、时间为 3.4h、pH 值为 5.22 的条件下,MSPs 的提取率最高(10.40%)。通过 DEAE-纤维素和 Sephadex G-100 层析,从锦葵粗多糖中分离得到 3 种均一多糖组分(MSP-1、MSP-2、MSP-3),其由不同摩尔比的半乳糖、葡萄糖醛酸、阿拉伯糖、鼠李糖和甘露糖组成,分子量范围为 2.6×10-8~8.8×10Da。这些组分能以剂量依赖的方式显著提高抗氧化、抗肿瘤和抗菌活性。MSP-2 显示出比 MSP-1 和 MSP-3 更强的抗氧化活性,包括还原能力和 DPPH 和 OH 自由基清除活性。MSP-2(1.0mg/mL)对 A549 和 HepG2 细胞生长的增殖抑制率分别为 45.1%和 53.2%。与革兰氏阴性菌(大肠杆菌 PTCC 1763 和鼠伤寒沙门氏菌 PTCC 1709)相比,革兰氏阳性菌(蜡状芽孢杆菌 PTCC 1015 和金黄色葡萄球菌 PTCC 1112)对各种 MSPs(3-15mg/mL)的敏感性较低。
