Pingmu Decoction is the Traditional Chinese Medicine which has treated Graves' Ophthalmopathy (GO) in the inactive stage for more than ten years. This study was to explore the mechanism of Pingmu Decoction of inhibiting preadipocytes in GO patients from differentiating into mature adipocytes. Human orbital preadipocytes were isolated and cultured through tissue explant method. Orbital preadipocytes were induced into mature adipocytes. The medicinal serum was prepared from rats. The cells were treated with medicinal serum which were divided into three groups, low dose group (5%), medium dose group (10%), and high dose group (20%). The cells viabilities were observed by Oil Red O staining, MTT method, and Annexin V/propidium iodide (PI) double staining. Effect of Pingmu Decoction on cell apoptosis rate of orbital matured adipocytes was measured by flow cytometry. The genes Fas and Fas L from cell groups were tested by RT-PCR and Western blotting. The expression of master adipogenic transcription factors, including peroxisome proliferation-activity receptor (PPAR) and CCAAT/enhancer binding protein (C/EBP) , was tested by Western blotting. Pingmu Decoction could reduce orbital preadipocytes viability and induce apoptosis of mature adipocyte via Fas/Fas L signaling pathway. Pingmu Decoction reduced lipid accumulation and downregulated the expression of PPAR and C/EBP . Pingmu Decoction may play a therapeutic effect by reducing the accumulation of orbital adipocytes.
平目汤是一种治疗Graves眼病(GO)非活动期已有十多年的中药。本研究旨在探讨平目汤抑制GO患者前脂肪细胞分化为成熟脂肪细胞的机制。采用组织块培养法分离培养人眼眶前脂肪细胞。将眼眶前脂肪细胞诱导为成熟脂肪细胞。制备大鼠含药血清。将细胞分为低剂量组(5%)、中剂量组(10%)和高剂量组(20%),用含药血清处理。通过油红O染色、MTT法和Annexin V/碘化丙啶(PI)双染法观察细胞活力。采用流式细胞术检测平目汤对眼眶成熟脂肪细胞凋亡率的影响。通过RT-PCR和Western印迹法检测细胞组中Fas和Fas L基因。采用Western印迹法检测过氧化物酶体增殖物激活受体(PPAR)和CCAAT/增强子结合蛋白(C/EBP)等主要脂肪生成转录因子的表达。平目汤可通过Fas/Fas L信号通路降低眼眶前脂肪细胞活力并诱导成熟脂肪细胞凋亡。平目汤减少脂质蓄积,下调PPAR和C/EBP的表达。平目汤可能通过减少眼眶脂肪细胞的蓄积发挥治疗作用。
