A Gram negative, aerobic, non-motile and rod-shaped bacterial strain designated as Gr-2 was isolated from granules used in a wastewater treatment plant in Korea, and its taxonomic position was investigated using a polyphasic approach. Strain Gr-2 grew at 18-37°C (optimum temperature, 30°C) and a pH of 6.0-8.0 (optimum pH, 7.0) on R2A agar medium. Based on 16S rRNA gene phylogeny, the novel strain showed a new branch within the genus Pseudaminobacter of the family Phyllobacteriaceae, and formed clusters with Pseudaminobacter defluvii THI 051 (98.9%) and Pseudaminobacter salicylatoxidans BN12 (98.7%). The G+C content of the genomic DNA was 63.6%. The predominant respiratory quinone was ubiquinone-10 (Q-10) and the major fatty acids were cyclo-C ω8c, C ω7c, and iso-C. The overall polar lipid patterns of Gr-2 were similar to those determined for the other Pseudaminobacter species. DNA-DNA relatedness values between strain Gr-2 and its closest phylogenetically neighbors were below 18%. Strain Gr-2 could be differentiated genotypically and phenotypically from the recognized species of the genus Pseudaminobacter. The isolate therefore represents a novel species, for which the name Pseudaminobacter granuli sp. nov. is proposed with the type strain Gr-2 (=KACC 18877 =LMG 29567).
从韩国一家污水处理厂使用的颗粒中分离出一株革兰氏阴性、需氧、无运动性的杆状细菌菌株,命名为Gr-2,并采用多相分类法研究其分类地位。菌株Gr-2在R2A琼脂培养基上于18-37°C(最适温度30°C)和pH 6.0-8.0(最适pH 7.0)下生长。基于16S rRNA基因系统发育分析,该新菌株在叶杆菌科假氨基杆菌属内形成一个新分支,并与脱氮假氨基杆菌THI 051(98.9%)和水杨酸盐氧化假氨基杆菌BN12(98.7%)聚类。基因组DNA的G+C含量为63.6%。主要呼吸醌为泛醌-10(Q-10),主要脂肪酸为环-C ω8c、C ω7c和异-C。Gr-2的总体极性脂质模式与其他假氨基杆菌属物种的模式相似。菌株Gr-2与其最接近的系统发育邻居之间的DNA-DNA相关性值低于18%。菌株Gr-2在基因型和表型上可与假氨基杆菌属已确认的物种区分开来。因此,该分离株代表一个新物种,提议将其命名为颗粒假氨基杆菌(Pseudaminobacter granuli)新种,模式菌株为Gr-2(=KACC 18877 =LMG 29567)。
