A Gram-stain positive, aerobic, non-motile, rod-shaped bacterium (THG-T1.18) was isolated from desert soil. Growth occurred at 20-35 °C (optimum 28-30 °C), at pH 5-7 (optimum 7) and at 0-4 % NaCl (optimum 0-1 %). Based on 16S rRNA sequence analysis, the nearest phylogenetic neighbours of strain THG-T1.18 were identified as Chryseomicrobium amylolyticum DSM 23442 (96.6 %), Chryseomicrobium imtechense JCM 16573 (96.3 %) and Chryseomicrobium aureum KACC 17219 (96.1 %). The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, two unidentified aminolipids and one unidentified glycolipid. The quinone system was composed of MK-7, MK-8 and MK-6. The major fatty acids were iso C15 : 0 and anteiso C15 : 0. The type of peptidoglycan was A4β, containing of l-Orn-D-Glu. The DNA G+C content of strain THG-T1.18 was 50.4 mol%. DNA-DNA hybridization values between strain THG-T1.18 and C. amylolyticum DSM 23442, C. imtechense JCM 16573, C. aureum KACC 17219 were 24.7 % (20.1 % reciprocal analysis), 19.5 % (16.1 %) and 10.4 % (6.7 %) respectively. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA-DNA hybridization data, strain THG-T1.18 represents a novel species of the genus Chryseomicrobium, for which the name Chryseomicrobium deserti sp. nov. is proposed. The type strain is THG-T1.18 (=KACC 18929=CCTCC AB 2016179).
从沙漠土壤中分离出一株革兰氏染色阳性、需氧、无运动性的杆状细菌(THG-T1.18)。该菌在20-35 °C(最适温度28-30 °C)、pH 5-7(最适pH 7)和0-4 % NaCl(最适0-1 %)条件下生长。基于16S rRNA序列分析,菌株THG-T1.18最近的系统发育邻种被鉴定为解淀粉金色微球菌DSM 23442(相似度96.6 %)、因泰克金色微球菌JCM 16573(相似度96.3 %)和金色金色微球菌KACC 17219(相似度96.1 %)。极性脂类为双磷脂酰甘油、磷脂酰乙醇胺、磷脂酰甘油、两种未鉴定的氨基脂类和一种未鉴定的糖脂。醌系统由MK-7、MK-8和MK-6组成。主要脂肪酸为异C15 : 0和anteiso C15 : 0。肽聚糖类型为A4β,含有l-Orn-D-Glu。菌株THG-T1.18的DNA G+C含量为50.4 mol%。菌株THG-T1.18与解淀粉金色微球菌DSM 23442、因泰克金色微球菌JCM 16573、金色金色微球菌KACC 17219之间的DNA-DNA杂交值分别为24.7 %(反向分析为20.1 %)、19.5 %(16.1 %)和10.4 %(6.7 %)。基于系统发育分析、化学分类数据、生理特征和DNA-DNA杂交数据,菌株THG-T1.18代表金色微球菌属的一个新物种,为此提出沙漠金色微球菌新种(Chryseomicrobium deserti sp. nov.)。模式菌株为THG-T1.18(=KACC 18929=CCTCC AB 2016179)。
