A novel aerobic bacterium, designated strain LAM9153, was isolated from a saline soil sample collected from Lingxian County, Shandong Province, China. Cells of strain LAM9153 were observed to be Gram-stain negative, non-motile, non-spore-forming and rod-shaped. The new isolate grew optimally at 30-35 °C, pH 7.0 and 0.5% of NaCl concentration (w/v). According to the phylogenetic analysis based on the 16S rRNA gene sequence, strain LAM9153 shares high similarity with Chitinophaga terrae Gsoil 238 (96.9%) and Chitinophaga niabensis JS 13-10 (95.9%), forming a subcluster with C. terrae Gsoil 238, Chitinophaga cymbidii R156-2, C. niabensis JS 13-10 and Chitinophaga soli Gsoil 219 in the phylogenetic tree. The major cellular fatty acids (> 10%) were identified as iso-C, iso-C 3-OH and summed features 3 (C ω6c and/or C ω7c). The predominant respiratory quinone was identified as menaquinone MK-7. The polar lipids consisted of phosphatidylethanolamine, aminophospholipid, three unidentified aminolipids and five unidentified lipids. The genomic DNA G+C content was determined to be 53.2 ± 1.6 mol%. On the basis of phylogenetic, chemotaxonomic and phenotypic data, strain LAM9153 is concluded to represent a novel species of the genus Chitinophaga, for which the name Chitinophaga salinisoli sp. nov. is proposed. The type strain is LAM9153 (= ACCC 19960 = JCM 30847).
从中国山东省陵县采集的一份盐渍土样本中分离出一株新型需氧细菌,命名为LAM9153菌株。观察到LAM9153菌株的细胞革兰氏染色阴性、无运动性、不产芽孢且呈杆状。该新分离菌株在30-35°C、pH 7.0和0.5% NaCl浓度(w/v)下生长最佳。基于16S rRNA基因序列的系统发育分析表明,LAM9153菌株与陆地噬几丁质菌Gsoil 238(96.9%)和尼亚宾噬几丁质菌JS 13-10(95.9%)具有高度相似性,在系统发育树中与陆地噬几丁质菌Gsoil 238、大花蕙兰噬几丁质菌R156-2、尼亚宾噬几丁质菌JS 13-10和土壤噬几丁质菌Gsoil 219形成一个亚簇。主要细胞脂肪酸(>10%)被鉴定为异-C、异-C 3-OH和总和特征3(C ω6c和/或C ω7c)。主要呼吸醌被鉴定为甲基萘醌MK-7。极性脂质由磷脂酰乙醇胺、氨基磷脂、三种未鉴定的氨基脂质和五种未鉴定的脂质组成。基因组DNA G+C含量测定为53.2±1.6 mol%。基于系统发育、化学分类和表型数据,得出LAM9153菌株代表噬几丁质菌属的一个新物种的结论,为此提出新名称盐土噬几丁质菌(Chitinophaga salinisoli sp. nov.)。模式菌株为LAM9153(=ACCC 19960 =JCM 30847)。
