As a kind of malignant tumor in the male genitourinary system, prostate cancer exhibits significantly increased occurrence. Prostate-specific antigen (PSA) expression can be seen in the prostate cancer, prostatitis, and other diseases, therefore, lack of diagnostic specificity. The miR-155 expression is abnormally increased in the tumors. Therefore, this study aims to explore the clinical significance of PSA combined miR-155 detection in the early diagnosis of prostate cancer.
A total of 86 patients diagnosed with prostate cancer were enrolled in this study. PSA and miR-155 gene expression in tumor tissue were detected by using Real-time PCR. The serum levels of PSA were measured by using enzyme-linked immunosorbent assay (ELISA). The correlation of PSA and miR-155 expression with age, body mass index (BMI), tumor volume, tumor-node-metastasis (TNM) stage, lymph node metastasis (LNM), and other clinicopathological features were analyzed, respectively.
Serum PSA expression and PSA gene in tumor tissue were significantly higher compared to that in adjacent tissues (p<0.05). PSA gene and protein increased significantly with the clinical stage of TNM and decreased following the increase of grade (p<0.05). The miR-155 level was significantly elevated in the tumor tissue compared with para-carcinoma tissue (p<0.05). PSA and miR-155 expressions were positively correlated with TNM stage, tumor volume, and LNM, and negatively correlated with grade (p<0.05).
PSA and miR-155 were closely related to the clinicopathological features of prostate cancer. Combined detection is helpful for the early diagnosis of prostate cancer.
前列腺癌作为男性泌尿生殖系统的一种恶性肿瘤,其发病率显著增高。前列腺特异性抗原(PSA)在前列腺癌、前列腺炎等疾病中均可表达,故缺乏诊断特异性。肿瘤中 miR-155 的表达异常增高。因此,本研究旨在探讨 PSA 联合 miR-155 检测在前列腺癌早期诊断中的临床意义。
共纳入 86 例前列腺癌患者。采用实时 PCR 检测肿瘤组织中 PSA 和 miR-155 基因的表达,酶联免疫吸附试验(ELISA)检测血清 PSA 水平。分析 PSA 和 miR-155 表达与年龄、体重指数(BMI)、肿瘤体积、肿瘤-淋巴结-转移(TNM)分期、淋巴结转移(LNM)等临床病理特征的相关性。
血清 PSA 表达和肿瘤组织中 PSA 基因均明显高于癌旁组织(p<0.05)。PSA 基因和蛋白随 TNM 临床分期的增加而显著升高,随分级的增加而降低(p<0.05)。肿瘤组织中 miR-155 水平明显高于癌旁组织(p<0.05)。PSA 和 miR-155 的表达与 TNM 分期、肿瘤体积和 LNM 呈正相关,与分级呈负相关(p<0.05)。
PSA 和 miR-155 与前列腺癌的临床病理特征密切相关,联合检测有助于前列腺癌的早期诊断。
