Abstract

The present study has been designed with the objective of determining if fatty acids bound to bovine serum albumin-V (BSA-V) can improve motility, viability, and increase acrosome reaction (AR) and utilization of glucose in boar spermatozoa. Boar spermatozoa were washed, swum-up and incubated at 37°C for 6 h in TALP medium supplemented with fatty acids bound to bovine serum albumin fraction V (BSA-V), fatty acid free BSA (BSA-FAF), polyvinyl alcohol + main fatty acids bound to BSA-V (PVA + FA) and PVA. Sperm motility, viability, AR, and the incorporation and oxidation of C-glucose were evaluated during 6 h of incubation. The results show that the BSA-V was superior to BSA-FAF and PVA in improving motility and AR. Viability was significantly increased ( < 0.05) by only BSA-V compared with PVA. When the main fatty acids compound of BSA-V were added to PVA, the sperm motility, viability and AR became almost the same as with BSA-V. The rate of incorporation and oxidation of C-glucose were significantly increased ( < 0.05) by BSA-V compared with BSA-FAF and PVA. Fatty acids bound to BSA-V are important for improvement of sperm functions. The present study postulates that fatty acids bound to BSA-V are important to acrosome reaction and the utilization of glucose in boar spermatozoa.

本研究旨在确定与牛血清白蛋白-V(BSA-V)结合的脂肪酸是否能改善公猪精子的活力、存活率,增加顶体反应(AR)以及提高葡萄糖利用率。公猪精子经洗涤、上浮处理后,在补充了与牛血清白蛋白V组分(BSA-V)结合的脂肪酸、无脂肪酸牛血清白蛋白(BSA-FAF)、聚乙烯醇+与BSA-V结合的主要脂肪酸(PVA+FA)以及聚乙烯醇(PVA)的台氏液(TALP)培养基中于37°C孵育6小时。在孵育的6小时内评估精子活力、存活率、顶体反应以及C-葡萄糖的摄取和氧化情况。结果表明,在改善精子活力和顶体反应方面,BSA-V优于BSA-FAF和PVA。与PVA相比,仅BSA-V能显著提高精子存活率(P<0.05)。当将BSA-V的主要脂肪酸化合物添加到PVA中时,精子活力、存活率和顶体反应几乎与使用BSA-V时相同。与BSA-FAF和PVA相比,BSA-V能显著提高C-葡萄糖的摄取和氧化速率(P<0.05)。与BSA-V结合的脂肪酸对改善精子功能很重要。本研究推测,与BSA-V结合的脂肪酸对公猪精子的顶体反应和葡萄糖利用很重要。
