Effects of Phytochemically Characterized Extracts From and Isolated Secoiridoids on Mediators of Inflammation in a Human Neutrophil Model.

Abstract

The aim of the present study was to investigate the effects of phytochemically characterized extracts connected with the traditional use (infusions and ethanolic extracts) of different parts of (common lilac) on the pro-inflammatory functions of neutrophils. Active compounds were isolated from the most promising extract(s) using bioassay-guided fractionation, and their activity and molecular mechanisms of action were determined. The extracts were characterized using a HPLC-DAD- MS method. The effects on ROS, MMP-9, TNF-α, IL-8, and MCP-1 production by neutrophils were measured using luminol-dependent chemiluminescence and enzyme-linked immunosorbent assay (ELISA) methods. The effects on p38MAPK, ERK1/2, JNK phosphorylation, and NF-B p65 translocation were determined using western blots. The major compounds detected in the extracts and infusions belong to structural groups, including caffeic acid derivatives, flavonoids, and iridoids. All extracts and infusions were able to significantly reduce ROS and IL-8 production. Bioassay-guided fractionation led to the isolation of the following secoiridoids: 2″-epiframeroside, oleonuezhenide, oleuropein, ligstroside, neooleuropein, hydroxyframoside, and framoside. Neooleuropein appeared to be the most active compound in the inhibition of cytokine production by attenuating the MAP kinase pathways. The present study demonstrated that common lilac, which is a traditionally used medicinal plant in Europe, is a valuable source of active compounds, especially neooleuropein.

本研究的目的是调查与传统用途(浸剂和乙醇提取物)相关的、以植物化学方法表征的丁香不同部位提取物对中性粒细胞促炎功能的影响。采用生物测定导向分级分离法从最有前景的提取物中分离出活性化合物,并确定其活性及作用分子机制。提取物采用高效液相色谱-二极管阵列-质谱法进行表征。使用鲁米诺依赖性化学发光法和酶联免疫吸附测定(ELISA)法测量提取物对中性粒细胞产生活性氧(ROS)、基质金属蛋白酶-9(MMP-9)、肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)和单核细胞趋化蛋白-1(MCP-1)的影响。使用蛋白质免疫印迹法确定提取物对p38丝裂原活化蛋白激酶(p38MAPK)、细胞外信号调节激酶1/2(ERK1/2)、c-Jun氨基末端激酶(JNK)磷酸化以及核因子-κB p65(NF-κB p65)易位的影响。提取物和浸剂中检测到的主要化合物属于不同结构组,包括咖啡酸衍生物、黄酮类化合物和环烯醚萜类化合物。所有提取物和浸剂均能显著降低ROS和IL-8的产生。生物测定导向分级分离法导致分离出以下裂环环烯醚萜类化合物:2″-表弗拉莫苷、橄榄新苷、橄榄苦苷(橄榄甙)、裂环烯醚萜苷、新橄榄苦苷、羟基弗拉莫苷和弗拉莫苷。新橄榄苦苷似乎是通过减弱丝裂原活化蛋白激酶途径抑制细胞因子产生的最具活性的化合物。本研究表明,丁香作为欧洲传统药用植物,是活性化合物特别是新橄榄苦苷的宝贵来源。
