This study hypothesized to evaluate the effect of betanin, a chromoalkaloid on plasma and altered tissues glycoprotein components in streptozotocin-nicotinamide-induced diabetic rats. Diabetes was induced by a single intraperitoneal (i.p.) injection of streptozotocin (45 mg/kg b.w.) dissolved in 0.1 M citrate buffer (pH 4.5) 15 min after the i.p. administration of nicotinamide (110 mg/kg b.w.). Experimental rats were administered betanin at the dose of 20 mg/kg b.w. and glibenclamide (600 µg/kg b.w.) once a day for 30 days. Diabetic rats revealed significant (p < 0.05) increase in the levels of glucose, HbA, hexose, hexosamine, sialic acid and fucose in the plasma; decrease in the levels of plasma insulin, Hb and sialic acid in the liver and kidney; significant (p < 0.05) increase in hexose, hexosamine and fucose in the liver and kidney. Moreover, periodic acid-Schiff staining of tissues revealed positive-stain accumulation in diabetic rats. On co-supplementation of betanin and glibenclamide to diabetic rats for the period of 30 days brought back the levels of plasma and tissues glycoprotein components. Based on the present study, we propose that betanin possesses significant protective effect on glycoprotein components in plasma and tissue of diabetic rats.
本研究假设评估甜菜红,一种色胺生物碱对链脲佐菌素-烟酰胺诱导的糖尿病大鼠血浆和改变组织糖蛋白成分的影响。糖尿病通过链脲佐菌素(45mg/kg b.w.)在烟酰胺(110mg/kg b.w.)腹腔注射后 15 分钟溶解在 0.1 M 柠檬酸盐缓冲液(pH 4.5)中单次腹腔注射诱导。实验大鼠每天给予甜菜红 20mg/kg b.w.和格列本脲(600µg/kg b.w.)一次,共 30 天。糖尿病大鼠的血糖、HbA、己糖、己糖胺、唾液酸和岩藻糖水平显著(p<0.05)升高;肝和肾中血浆胰岛素、Hb 和唾液酸水平降低;肝和肾中己糖、己糖胺和岩藻糖水平显著(p<0.05)升高。此外,组织过碘酸-Schiff 染色显示糖尿病大鼠的阳性染色积累。甜菜红和格列本脲共同补充糖尿病大鼠 30 天,可恢复血浆和组织糖蛋白成分的水平。基于本研究,我们提出甜菜红对糖尿病大鼠血浆和组织糖蛋白成分具有显著的保护作用。
