A novel actinomycete, designated strain YIM CF22, was isolated from a desert soil sample collected from Turpan in Xinjiang Uyghur Autonomous Region, north-western China. The taxonomic position of the strain YIM CF22 is described based on a polyphasic approach. Strain YIM CF22 was found to form irregular sporangia on agar media. It contains meso-diaminopimelic acid in the cell wall peptidoglycan. The major menaquinone was identified as MK-9(H); the polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, two unidentified phospholipids and two unidentified glycolipids. The whole cell sugars were found to be ribose, mannose, galactose, glucose and xylose. The major cellular fatty acids were found to be (> 5%) iso-C (43.5%), anteiso-C (10.2%), iso-C (7.1%), C ω8c (6.3%) and iso H-C (5.9%). The G+C content was determined to be 70.8%. 16S rRNA gene sequence analysis of strain YIM CF22 showed high similarity (97.0%) to Actinoplanes rishiriensis NBRC 108556. The strain also showed high 16S rRNA gene sequence similarities to Verrucosispora sediminis CGMCC 4.3550 (96.9%) and Micromonospora tulbaghiae DSM 45142 (96.8%). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain YIM CF22 clusters with A. rishiriensis NBRC 108556, Actinoplanes globisporus JCM 3186 and Actinoplanes rhizophilus NEAU-A-2. Based on the differential phenotypic characteristics and the results of DNA-DNA relatedness and phylogenetic analysis, it is proposed that strain YIM CF22 represents a novel species of the genus Actinoplanes, for which the name Actinoplanes deserti sp. nov. is proposed. The type strain is YIM CF22 (= KCTC 39543 = CCTCC AB2018113).
从中国西北部新疆维吾尔自治区吐鲁番采集的沙漠土壤样本中分离出一株新型放线菌,命名为YIM CF22菌株。基于多相分类法描述了YIM CF22菌株的分类地位。发现YIM CF22菌株在琼脂培养基上形成不规则的孢子囊。其细胞壁肽聚糖中含有内消旋二氨基庚二酸。主要甲基萘醌被鉴定为MK-9(H);极性脂被鉴定为双磷脂酰甘油、磷脂酰乙醇胺、磷脂酰肌醇、两种未鉴定的磷脂和两种未鉴定的糖脂。全细胞糖被发现为核糖、甘露糖、半乳糖、葡萄糖和木糖。主要的细胞脂肪酸被发现为(>5%)异-C(43.5%)、anteiso-C(10.2%)、异-C(7.1%)、C ω8c(6.3%)和异H-C(5.9%)。G+C含量测定为70.8%。YIM CF22菌株的16S rRNA基因序列分析显示与立尻游动放线菌NBRC 108556具有高度相似性(97.0%)。该菌株与沉积疣孢菌CGMCC 4.3550(96.9%)和土生小单孢菌DSM 45142(96.8%)也显示出较高的16S rRNA基因序列相似性。基于16S rRNA基因序列的系统发育分析表明,YIM CF22菌株与立尻游动放线菌NBRC 108556、球形游动放线菌JCM 3186和嗜根游动放线菌NEAU-A-2聚类在一起。基于不同的表型特征以及DNA-DNA相关性和系统发育分析结果,建议将YIM CF22菌株代表游动放线菌属的一个新物种,为此提出沙漠游动放线菌新种Actinoplanes deserti sp. nov.。模式菌株为YIM CF22(=KCTC 39543=CCTCC AB2018113)。
