Oncolytic viruses (OV) are engineered to infect, replicate in and kill cancer cells. Currently, the OV therapeutic approach is mainly restricted to neoplasia amenable to direct local administration of viral particles, while the possibility of a systemic delivery of cancer-tropic viruses would extend the OV application to the treatment of metastatic neoplasia. Herein, we applied imaging to demonstrate that cancer tropism is achieved when OV are encapsulated inside extracellular vesicles (EV) administered intravenously (i.v.), but not when injected intraperitoneally (i.p.). Moreover, we show that the therapeutic procedure adopted does not alter the immunomodulatory properties of the viruses.
溶瘤病毒(OV)经过基因工程改造后,能够感染、复制和杀死癌细胞。目前,OV 的治疗方法主要局限于可以直接局部给予病毒颗粒的肿瘤,而能够全身递送肿瘤趋向性病毒的可能性将会扩大OV 在转移性肿瘤治疗中的应用。在此,我们应用成像技术来证明,当OV 被包裹在细胞外囊泡(EV)中并通过静脉内(i.v.)给药时,OV 能够获得肿瘤趋向性,但当通过腹腔内(i.p.)注射时则不能。此外,我们还表明,所采用的治疗程序不会改变病毒的免疫调节特性。
